I got chills. This whole conversation was amazing. ("Your words are as empty as your future" remains, to this day, my single favourite line out of the whole two games)
Oh, and yes, the opening scene of ME1. Also, Saren blowing his brains off as a result of Shepard's persuasion. Wow.

ME1 Sovereign reveal: *jaw dropping to floor* "...this will be an ...epic... story"
ME2 Being rebuffed by Miranda on Minuteman station: "I like this woman."
The combination of the Reaper plot and my most favorite video game character of all time, that's what made the ME games special for me. Ah, and that it's a sci-fi universe. Fantasy worlds are a dime a dozen, good sci-fi universes rather rare. And for the standards of visual media, the ME universe is a rather good one.

- Virmire and the Sovereign reveal

Pretty much this for me as well, except the part about buying a 360 for it, as I was already xbox-indoctrinated at that point. But I was convinced enough that the game would be great from those early trailers that it was pretty much my "must play" title for ages before it came out.
In terms of actual game moments: Walking into the Citadel Presidium for the first time. I misss my ME citadel in ME2 :-(

The explicit references to his identity were cut, however. They had different lines depending on whether or not you just talked to him, punched him, or never even opened the door to the shed.

Sees Mass Effect, looks at the backside descriptions: "I think I read something about this one, don't know though, think I remember it was good game as far as reactions were... hmm"
Buought Mass Effect -> Went home again.
Whilst Mass Effect installed, I read the manual: "That really does look like a nice one. May just have been the most lucky I'm-bored-buy of time for me..."
Installation has finished: "Want to start the game?" -> Yes
Game starts ... then I hear Vigil and see the opening menu -> This is it.
Other moments like that in order of greater magnitude are:
End of Eden Prime: This game isn't just good, it's very good!
First time on the Citadel: This game is getting even better and better, damn!
Virmire: HOLY MUTHA****IN' SHIZNIT!?!? This is the best game I have played!
Everything after Virmire (played the entire endgame in one go (yeah, it was THAT good) even though it was already 2am in the morning only cemented my opinion.
