If I have one user on the PS3 and on that user I have 2 seperate playthroughs saved, when I get the DLC is there a way to apply the DLC to both of those playthroughs so each one has the extra characters and missions. If so, How? Please and thank you.

yes just load up the saves and the DLC will be active.
