They will already be on (if you want to turn off press shift+caps lock for gameplay version, shift+F12 for everything else. Press Pause/Break key to turn off antialiasing).
EDIT: Looks like it's not possible to use the injector to circumvent the Origin overlay issue:

Are you using anything like XFire or another game overlay program? For screenshots, I'm used a modded coalesced.bin with UI hiding and flycam. It will be included with the mod release this week.

Answer: neither T_T. Just finished recording in-game footage. My laptop can't handle the AO @1080p haha, I had to slow down the game by 3x to get smooth footage

EDIT: I've been getting some comments lately about how this mod doesn't look "different" enough from stock. It's all good feedback, but instead of responding to individual posts, here's my reasoning for all to see
Once you start creating something for the sake of making it different, it's not about making the experience better - it's about making it different just for the sake of being different. Stock ME3 is quite good (as I have mentioned several times below), but I think that at times, the color palette is too saturated, considering it's a war story. As such, I've toned down the saturation, added a bit of ME1 blue, and tweaked the bloom (barely. Toggle bloom in-game with shift+scroll lock to see what I mean).
If you guys want something different, then I'm more than happy to provide an ENB that does just that. In any case, I just wanted to get my reasoning across. Thanks for all who have tried the mod - I appreciate it!

Mass Effect 3 Cinema Mod [Gameplay Optimized Ver][/b]
This version is optimized for gameplay. It has the following features:
- Specific options toggle for bloom, color correction, and SMAA (all can be turned on/off in-game via hotkeys).
- Good performance. Only ~10fps hit in performance. Even less with Bloom turned off.
- Color correction Imitates human eye. This means shadows have a (very) slight blue tint. Looking into a very bright light source desaturates colors on the screen dramatically (a lot of people don't like the bright bloom from lights, so I chose this solution instead).
- Highly customizable (like all ENB-based mods).
- Easy install. Just copy files to your ME3 install dir.
- Works with Mass Effect 1, 2 and 3 (but designed specifically for 3).

This is a WIP version of my Mass Effect 2 ENB config ported to the Mass Effect 3 demo.
This mod is not for everyone! Default ME3 graphics are quite good, but sometimes it's nice to see things from a different perspective. This mod intentionally sacrifices some clarity and IQ in shadowed areas in order to create a more dramatic and cinematic effect. A blue filter is also applied to add a colder, more foreboding atmosphere (remember, this is the galaxy at war!).
This mod is a work-in progress and will be tweaked for the final release of the game.
CURRENTLY, YOU MUST DISABLE THE ORIGIN OVERLAY FOR THIS TO WORK.
To do this, open Origin, click the gear icon in the top-right. Then go Settings > In-Game > Uncheck "Enable Origin In-game". If you do not do this, there is a good chance that ME3 will crash when using this mod.
To Enable/Disable the effect in-game
Press Shift+F10 to disable bloom. Shift+F12 to disable color correction + Bloom. Pause/Break to toggle SMAA.

What does this mod do?
- Tweaked colors to better follow what the human eye perceives IRL.
- Shadows are darker and bluer.
- Overall bloom is toned down, however, intense light sources (such as the sun) have a more focused bloom.
- Colors in shadow are desaturated, whereas highlighted colors are saturated.
- Ditches in-game FXAA for more efficient SMAA algorithm (which also improves IQ, imo).
- EXPERIMENTAL film grain (not very good yet - not really like ME1/2 film grain)
- On my laptop (core i7, nvidia gt 330m), I lose about 10fps - from 50fps to 40fps. You can expect a similar performance cost, however, the game remains perfectly playable.

Instructions to enable Ambient Occlusion on machines with Nvidia graphics cards:
1) Download Nvidia inspector:

2) Extract Nvidia inspector to it's own directory and run the .exe (nvidiaInspector.exe)
3) Click on the wrench/screwdriver icon in the mid-right, next to the Driver Version info.
4) In the window that opens, click the dropdown menu next to Profiles, and scroll down to Mass Effect (type m to quickly get to the entries that start with m)

5) Check to see that Ambient Occlusion compatibility is set to 0x00000020 (Mass Effect). Then, click the "Add application to current profile" icon (it's the one right next to the big, red X).

6) In the dialog window that opens, navigate to your Mass Effect 3 Demo\\Binaries\\Win32 directory and select MassEffect3Demo.exe. Once you do this, you should see the ME3 exe added to the green bar at the top

7) Hit apply, and you're all set! Launch ME3 from origin, or directly via the ME3 executable.

- Standalone Film Grain should actually be standalone now (no more darkening).

Ah...FINALLY found it. Virtually all the "download" buttons on the pages are NOT for Inspector, though they look like it. They are all for CRAP like various download managers, music downloaders, etc,etc. It takes some hunting to find the REAL download link for Inspector.

You need an external program like NVIDIA Inspector to enable AO in the game. By default, the NVIDIA Control Panel has the Ambient Occlusion setting for Mass Effect 3 but it's greyed out, hence the need for an external program.DVACDK wrote...

ENB alone will not enable AO. At least not for ME3. It does for other games where ENB was designed for AO, such as GTA: San Andreas. But if I recall, the AO was inaccurate as hell (sometimes the AO effects would be created around HUD objects!)
It's weird, ME3 clearly supports Ambient Occlusion, but for some strange reason, Bioware or NVIDIA decided to have it permanently disabled except for modders like us.
