Bioware mentioned that playing multiplayer assists you in the single player game by adding to your "readiness" score. Does this mean specifically that playing multiplayer can replace any and all planet probing entirely and still allow players to achieve "the best" ending per se?

Yes.

But from what I remember, multiplayer maps/missions are unlocked via single player probing. I don't think your readiness score will go up much if you do the same mission over and over again.
