Just wondering if the disc for Mass Effect 3 will need to be in my computer whenever I play, like ME2, or do I not need it after the installation.
(I am aware that I can always type the product key into origin and download the game that way instead, but it'll take several hours for me to download the game.)
Is the disc required to play mass effect 3?

since you need Origin Registration, you don't need the disk in order to play.

Clients like Origin and Steam negate the need of having the disc in the tray.

No, but the Origin client must be installed to play (even for retail copies). The Origin client will launch automatically when you start the game (the discs will never be required past the initial installation).
