I am running Windows XP SP3 and had no problems playing the Mass Effect 3 demo.
Now when I'm trying to pre-load the game I get an error message that my Operating System is not supported and that I need to upgrade to a newer version of Windows to play the game.
Considering that the requirements are Windows XP SP3/Vista SP1or Win 7, and as mentioned above, the demo ran just fine, why am I getting this error?
It would be greatly appreciated If anyone could help with some information.

I'm also getting the same message. I assume its an error as the OP stated XP SP3 is winthin minimum requirements.

I've actually heard that antivirus programs can cause this problem. What antivirus are you running?

Disabling antivirus software didn't help with the issue but thank you for the suggestion. I'm hoping EA puts a fix in quickly.

Seeing two of you folks within minutes of one another makes me feel very good, and if the harsh-sounding announcements of tougher enforcement (the ToS really covered it already, for the most part, but just wasn't enforced rigprously) are what it takes to see your enthusiasm, then it's done its job, very pleased to have you among us (now, can we please get some kind of FLAG set for those of us who have read the thing a thousand times already, to give us a little relief?)

Hi. I have almost the same problem but reinstallation doesn't seem to cure it. I get the error even when starting from Origin (16bit incompatibility) or desktop shortcut. I tried uninstalling Origin and ME3 and then reinstalling. I then tried updating the freshly downloaded ME3, it pulled in another 40MB-ish, which seemed odd, since I've only just downloaded the main file! It made no difference, nothing I've done seems to resolve the issue. This is a Win7 64 machine that had no trouble with ME2.

Thanks for getting that update through so fast, I'm able to get to the release date check. Can't wait for 9 o'clock tonight!

Because this thread has actually had a response from Bioware, I would like to add to the "16-bit incompatibility" issue:
Since Sunday I have been trying to run the pre-loaded installation. As with previous Origin games, I fully expected to see the "Release Date Check" screen pop up...unfortunately, this did not happen. When trying to run the game from the Origin launcher, nothing happens. I get the Windows is busy mouse pointer for about two seconds, then nothing else happens. Even Task Manager is devoid of any ME3 or Origin activity (outside of the Origin.exe process). So, I browse into the ME3 install folder and run the MassEffect3.exe file directly, whereupon I see the error message "Unsupported 16-bit Application". Incidentally, after running the exe, the file resizes down to approximately 21MB and subsequent attempts to run the file result in a different error message (I cannot recall the exact wording). At this point, if I run "Check for Update" for ME3, it will download the 40MB file. If I have not attempted to run the exe, Origin comes back with a "Game is up to date" message and nothing is downloaded.
Steps I have taken to try to remedy this issue:
1) Check for Update (to ME3)
2) Repair installation (of ME3)
3) Uninstall Origin, download the newest version directly from the Origin website, reinstall
4) Uninstall ME3 and redownload/reinstall the game
5) Run the offending exe in compatibility mode
6) Run the offending exe in administrator mode
7) All combinations of the above
So far, nothing has fixed the problem. Now, I have seen it repeated over and over that this won't be an issue since the release date hasn't even passed...except that I (and, from the sounds of things, a lot of other people) cannot even get to the Release Date checker application. Even running the Release Date check application directly from its exe results in nothing happening. Is there any other potential fix that I have neglected to consider?
EDIT - My apologies, forgot some potentially pertinent information:

For what it's worth, I had this exact problem last night. I rebooted in frustration, then ran a Repair Installation on ME3. (Right-click on the game poster in Origin, and select Repair.) Origin sat there for a while with a progress bar, then claimed everything was fine. I clicked on the Play button one more time without expecting any change... and hello, there's the unlock screen.

Origin is a completely broken platform, and Bioware should release a patch allowing players to bypass origin completely.

Can you please try uninstalling Origin and then downloading the latest Origin client from here and see if the problem persists?
Thanks
I have done this, twice. The problem persists. The only thing ME3 releated I've been able to do in the last two days is read these forums and close the "Mass Effect 3 has stopped working" windows. which make me laugh, becuase as far as I can tell the game doesn't work at all.

Hi from Italy.
Here I'm having the same issue as a lot of users, for what I can see on :"The program or feature "\\??\\C:\\Program Files (x86)\\Origin Games\\Mass Effect 3\\binaries\\Win32\\MassEffect3.exe" cannot start or run due to incompatibility with 64 - bit versions of Windows. Please contact the software vendor to ask if a 64 - bit Windows compatible version is available." (english version of my italian error).
Tried: uninstall-reinstall of ME3, uninstall-reinstall of Origin, origin cache cleaned, windows TMP stuff cleaned, registry cleaned, antivirus deactivation, and everything else that has already been suggested here.
Some more information:If I DELETE masseffect3.exe and run the repair installation tool, it will correctly download it again. The downloaded file is 40,1 MB (42.078.464 byte), and has a shiny icon of ME3.
If I try to run it (both from origin or explorer) the file become corrupted:21,3 MB (22.370.035 byte), and the standard anonymous windows app icon.
I can send both files to anyone at EA/Bioware if asked to, with anything else you ask for.
Now, seen that I keep my pc and windows smooth and running (I am not the average gamer, I'm a senior computer analyst with 10+ years of experience of ANY environmente), that no other program/game has ever presented any problem and that a lot of other users out there have the same problem, can you suggest anything BUT not to reinstall windows?
That would be an offence to my intelligence, and a example of a bad technical support.
