I got the same issue as most of people.
I got it on DVD but my player doesn't work now so redeemed my cd-key and downloaded ME3 via Origin. I installed all the updates then pressed play. Nothing happened. The game shows on my task bar, seems as if it's thinking but then nothing happens, no error or anything like that. When I click on on on the taks bar a tiny window appears but then I get the message that the program doesn't respond. I've reinstalled the game twice and did the same with Origin. I've tried all the hints reccomended in here. Running Origin and the game as the admin, disabled my antivirus, closed all other programs, copied awc.dll file from the link. It still doesn't work.
I had the same issue with the demo. Sometimes it worked, sometimes it didn't. I've been reinstalling it a few times and after many tries it worked, just to stop working the next day.
But I'm too tired to download the game for the forth time - it takes 5-7 hours to dowload.
I have Windows Vista 32bit and nVidia GeForce 9800GT
Intel®Core2 Quad Q8200 @ 2.33GHz, 4GB RAM
I also copied EACore.dll

Do you use Microsofts EMET by any chance or had someone secure your setup/ system as much as possible?
Aside from that getting in the way (which might be possible, as it had been in the past with all kinds of DRM even Microsofts own infamous "Live") I can only think of the 2 files being copied to the wrong folder (if you do a clean install without backing up anything you would get a prompt to replace the 2 existing dll files).
On the "downloading the game again" part, "Origin" aids/ handles the installation from DVD, it might look like it's downloading the game through Origin when you launch the Installer on the DVD when in fact it starts the installation but from the 2 DVD's although through the Origin program (as intended), some ppl think it's downloading the game, cancel the setup and end up downloading the game later through Origin manually (without the DVD's), understandably tho as it might not have been clear enough.

It's not about being a multi user (account) thing, just thought that if you had someone secure the system for you he might have used Microsofts EMET to harden security which might cause issues.

(custom document/ system folder locations can also cause issues; which again can arise if someone setups a system with hardened security or other reasons)

First
make sure you have a documents folder in your Win7 profile, top right
of start menu. Also make sure you have no problems with your 'Personal'

I repaired my documents folder link (just having a folder called my
documents or adding it to the documents library isn't enough) I could
use the DLC.
If you did have the DLC working once, then that is a
different problem. This is about day 1 DLC problems before you start
the game.
Also I am glad I made codes for both my 1st and 2nd shepards, as it didn't pull across their looks.
The ME2 codes do work, dunno if you can figure out the code from a new game plus in ME2?
Also it seems sometimes only the Beta Version of Origin might be able to verify some of the ME3 DVD's, and thus start the installation from disc:
