It started working again after I uninstalled and re-installed origin and repaired the game in origin.

Yeah. I reinstalled Origin and it started working again. How do I repair the game through Origin though? I am having another issue and I don't see a repair thing anywhere.

Go to Mass Effect 3\\Binaries\\Win32 right-click on MassEffect3.exe and check 'run as administrator'. It worked for me. (And run Origin as an administrator)

Unfortunately Ilanox's solution does not work for me.
I used to be able to bypass this problem as it would arise after I had succesfully started the game. Then Origin would not close, so I would just simply close it in the Task Manager then play.
As of now, ME3 and Origin will not launch. I have tried the solutions listed in this forum. Is there something we need to do for an official to recognize this thread?

did you try to shut down the anti-sirus down then start the game ?

This is happening to me now, Origins updated, I try and start ME3 and I get a brief black screenthen it just crashed to windows. I've tried several solutions presented on the forums I've repaired ME3, updated graphics drivers etc. and nothing has helped.
