"Priority" missions, despite the name, are the ones you want to save for last if you want to get the maximum amount of content/game play. Priority missions will advance you from one Act to the next eventually, and not all sidequests are valid in an act different from the one you got them. I'm not sure which ones though. This also might apply more to the divide between Act II and Act III.
