Obvious spoilers below.
I don't believe the next game will be a continuation from Mass Effect 3. There are just too many decisions for Bioware to work with.. the variables have overwhelmed and boxed them in to the point where they are just killing everybody off.
You can see it with ME3.. Pretty much every Mass Effect 2 squadmate can die. I don't think they're going to devote much more resources to people that aren't there for 50% of people. Bioware cannot further elaborate on the choices the way we expect. If they are mentioned at all in the future they'll be reduced to side quests and one-line remarks.
So, the important question is.. are they going to work for hours, days, months on paths many people didn't take?
Are they going to further develop characters that were killed in a lot of playthroughs? I don't think so.
So what can they do? a prequel? I think that's possible..
Or....
Will we be re-living the events of Mass Effect 3 from a different perspective?
My prediction: We saw none of Earth except for the beginning and end of the game. Relatively small parts. What if we experience the fight on Earth from the perspective of the Big Ben Sniper seen in the announcement trailer? Obviously a lot went down that we did not see. But I wonder whether or not that was on purpose... They may not make a whole game out of it. But maybe an expansion pack or something.

I hate prequels.
I wouldn't mind a game focused on the Raloi a while after ME3 takes place and their expansion into the galaxy. Or something.

Yeah. This prediction isn't necessarily what I want.. more of a likelihood to what could come next.

Not doing DLC that doesn't give me alternate endings. Sorry. DLC should actually CHANGE things. See: Lair of the Shadow Broker, Arrival.

Since the Franchise is pretty much dead from the last part of ME3 onward, it's going to be rough. If it is a setup for a MMO like some are suggesting then they are going to have to deal with the fallout of turning a trilogy into a giant commercial.
But that said, I wantted some DLC like retaking Omega with Aria. I read the comics, stupidly, and I want to see how she kicks the remains of Cerberus off there. Of course if it takes place after the destruction of the relays then they will simply starve to death...
