I think you need to sneak next to them in cover and then an Icon will appear. All you have to do then is push the button you push for everything else (spacebar) and he should do a quick kill. TBH though I never really use it.

Basically, the enemy has to be on the other side of the cover, instead of the arrow that indicates Vault, a fist should appear. You press the melee button and BAM!
This is useful, since you can kill enemies with full shields. Just don't try on Phantoms, learned the hard way not to do that,
