Easily, Legion's final lines during the end of the quarian/geth arc. I remember distinctly when he asks the question, "Does this one have a soul?" I couldn't even think for a moment.

I like playing Renegade Shepard and never feel bad about my decisions but shooting that innocent Salarin in the back and then seeing him try to reach that console to release the Genophage cure really made me feel horrible. That moment to me was so powerful and I can't imagine how much worst it would have been if I had killed Mordin instead.

Rethinking about it, the scene where Anderson and Shep are stitting on the dias area, Anderson gives his "You did good, child" speech, dies and the amazing piano score makes that a perfect end point.

Most powerful? Definitely the destruction of the Quarian race, then Tali's suicide, then Legion's death - if you sided with the Geth on Rannoch. Definitely the largest and most devastating story moment...

Most powerful for my playthrough was the argument between Joker and Shepard on the bridge after Thessia falls. There is just so much raw anguish in Mark Vermeer and Seth Green's performance. I've rewatched that scene half a dozen times.

All the ones mentioned are good - some awesome but the one that spoke to me was Rila's sacrifice.
