It's a conversation option after you speak with his sister, the one about "everyone being out for themselves" or something along those lines.

But where and who is his sister?

You need to trigger Alistair's personal quest first, where he asks you if you have time to look for his sister if you're ever in Denerim. I'm not sure, but I think you also need to know the truth about his heritage for the quest to trigger.

I believe that you have to finish the Broken Circle quest first. Either that or you need to have high enough approval with him. He has to talk to you about it to get the quest, and after talking to his sister you have to choose the right dialogue line to harden him.

For me, he first talked about his sister after the conversation you get when you enter Redcliffe with him. A little chit-chat about his 'bastardness', a little chit-chat about something else, then boo, here you go.

You will find hs sister in Denerim, Market District, one of the houses will be marked by her name.

He needs to mention that he's a bastard before he tells you about his sister. I can't remember if you need to know that he's Maric's son or not yet, but once he's told you his mother was a maid, next time you go to Denerim with him, he'll bring up his sister. Go towards Wade's Emporium, and Alistair should point out his sister's house.

Hmmm.. I'm ready to go to the landsmeet. He has never mentioned his sister. No dialogue options are available with him. However my approval with him has never been stellar.

Then you likely won't get the quest to meet his sister and therefore harden him. He has to trust you at least a little to tell you about his family voluntarily.

Get his approval up and go redcliff then he will say some stuff about him being a royal bastard after if you work on his approval when you next speak in camp he will say he has a sister in denerim

Intersting. I finished gathering allies before starting the landsmeet so I went to denerim late in my game cycle. He mentioned his sister in camp so I took him on purpose. Don't remember the exact dialog unfortunatelybut after that I noticed that he seemed more macho. He took the kill blow on the traitor (no, I did not opt for anothe rgrey warden) and gave a decent speach before the final battle. But maybe it was more my imagination than an actual change.

You can get his personal quest after you enter Redcliffe. If you put Alistair on the throne he'll give the final battle speech. Toughening him up just makes it easier to convince him to be king.

All before Landsmeet:
Go to redcliffe. Then, get his approval up. Talk to him until he mentions seeing his sister. (Sometimes, he'll come to you and start the convo. Not sure of the conditions.) Go to Denerim, market district (first place you go from outside of Denerim). Walk along the North Side of the Market Square with Alistair in tow. He'll say that there's his sis' place. Go in, do the dialog. Outside, tell him that some people look out for themselves. Later, talk to him and confirm that he, too, should look out for himself. Voilá! Hardened Alistair is ready to take the throne with you in tow as an adviser or wife.
