If I incresae the readiness through multiplayer will it effect all of my single player characters?
I'm wondering because I wanted to do a minimal run to see what happens and I don't want anything increased or as little as possible. If multiplayer will effects this, then I'll hold off on playing it.

Yes. Since it is separate from your single player game, its results are applied to all of your saves. Same goes for promoting a multiplayer character.

One time I got disconnected while playing single-player or before loading can't remember but my readiness went back to 50%. I just tried playing offline again but it didn't go down... interesting

I also read somewhere that there is a rate of decay. So if you stop playing multiplayer it slowly decreases. Is this true?

Yes it decreases over time, at the moment I don't know by how much it does so per day. I've been playing a good bit of MP the last few days and the decrements of my galactic readiness when I start up the game seem to be almost random.

It slowly drops over time, but it's very quick to replace so I wouldn't overly worry about it. Jump onto MP for a quick run before you intend to hit the final SP mission and you'd get it up around 100% without much work.I also read somewhere that there is a rate of decay. Played a little bit of multiplayer a couple of days ago just to make sure my EMS would reach 4000, I got my galactic readiness level up to 60% and since then have play single player offline and it hasn't dropped.

Aha, thanks for the info! Seems like the GRL is managed by the servers. Dissallowing ME3.exe to connect could thus be a good idea to complete a new playthrough without intermediary MP to keep the GRL high.
