Sorry but this is incredibly unrealistic. Who decides 'the most important ones'?
The reaction to DAO's character-background DLC wasn't exactly golden, and that team said they'd probably move to packs that used the main character.

They can still get it done in a span off time it takes to make the next mass effect game. One DLC can contain at least 2-3 characters and so on and so on. I'd be willing to wait months for those to be relased at a time.

I wouldn't be mad at this but even if they did that first later down the line I think they should release something for garrus and ashley. Don't really care for liara so we can skip her.

-> Jack: on a mission with Miranda.
-> More war assets related to those five guys.
I'd like to see something like that.
- "Survivor Pack" - contains both Virmire Survivors
-> Special series of missions Alliance/VS related. Re-introduction of the Mako.
-> Revised & expanded romance arc
-> Additional war assets
-> Additional non-romance "friendship" path
... yeah. Guess something like that. Each pack should add another 3 or 4 hours of gameplay at least.

Jacob is the least "like" character among crew.
So.... no... I don't think it would work.
And for me? Miranda, Thane and Zaeed - yea, I would love to (aside from Garrus of course). The rest? I don't care much for them.

VS is already second the most developed companion in the game and have second best romance and you want exclusive DLC for him, but all other former companions that are now completely unimportant minor NPC should only get crappy all-in-one dlc?
I know that everyone want to get more of their favorites but, come on.

And still - if you compare the VS with Liara, you suddenly realize why I'm talking like that.
Everything that makes Liara's romance that fulfilling and enjoyable is missing for Ashley and (on a slightly lesser extend) Kaidan.
I just told this a friend via PM: the VS romance feels incomplete since the illusion of the VS character gets damaged by lack of interaction OUTSIDE the romance. Check out Liara: you get plenty of interaction with her in the game, even if not romanced. Same for Garrus and Vega - I'm happy BW still has the skills to create amazing characters.
But with the VS, they failed. If you take away the romance arc, you only have two extra characters - and redundant ones too. Kaidan can be replaced by Liara. Ashley can be replaced by Vega or Garrus.
So all VS has is the romance arc. And nothing else.
Which is the reason why I want more content for them. Give'm some love, give them Liara!treatment.
And while BW is at it, they also should do some great justice for ME2 chars too. I doubt they're going to make a $10 DLC for each ME2 character - and if they do, it might feel like a scam.
No need to concur here, it's just my opinion.

An attempted coup needs to be put down on Tchunka. Certain rival Turian factions don't like the new primarch and need to be taken care of. Shepard has to work with his/her mom or other human captain on a mission. Shepard needs to rally members of his/her old street gang (or civilians in general) as part of the human resistance movement (featuring Anderson). Or Shepard could find a Cerberus operation that took part in Akuze. Batarian slavers figure they're doomed anyway, and want revenge on Shepard before they die by avenging Torfan.
