Leliana: While confronting Marjoline, make sure you encourage Leliana to kill Marjoline rather than let her go. Maroline herself is a boss level character with two mages on either side. Afterwards, back at camp, Leliana will talk about how she feels regarding Maroline. Make sure you chose the options that make her believe that killing is a part of who she is.
Alistair: After his conversation with his sister, tell Alistair that people are looking out for themselves. Back in camp, tell him that he needs to stand up for himself.
This is essentially correct, but I'm going to add a couple of extra things you should know - and point out one error.
First the error, you don't have to have Leliana kill Marjolaine. You can threaten Marjolaine and make her stay away from Leli without killing her (I've done it about 5 times now). Simply choose conversation options back at camp with Leliana that tell her you think she was 'hiding' in the chantry, and that she wasn't meant for the convent life. Of course, if you want to kill Marjolaine, go ahead - but it's the conversation choices that 'harden' her, not the death. If you kill M. and then tell Leliana that she can choose who she is, doesn't have to be like M. and so on (the 'soft' choices) then Leliana won't be hardened.
With Alistair, if you 'harden' him he will nominate himself for king. If you are female and are romancing Alistair, you may want to consider your options on this one. If you harden him, he will nominate himself for king and this may cause complications depending on various factors.
If you are a Human Noble, and plan on setting Alistair up to rule with yourself as consort, go ahead.
If you are not a Human Noble and Alistair becomes king he will dump you - or keep you as his lover if your 'persuade' is high enough. So, if that's okay with you, again, go right ahead. If, OTOH, you would like your Grey Warden and Alistair to spend the remainder of their lives together, you may prefer that Alistair not become king. In this case, do not harden him. And when you choose to support Anora, remind him that he told you he didn't want to be king. In other words, you're saving him from a ghastly fate.
