Yes, this moment. This was probably the first time I honestly had a good hearty laugh in a video game's singleplayer. Whoever did the face animation for that cutscene, I want to shake that person's hand. I was worried that when EDI became a squadmate that she'd just be a boring robot trying to find humanity (like Data from Star Trek), but they did a real good job on making her mesh with the crew and giving her moments like these.

Another moment that really caught me off guard was the bromance scene with Garrus where you're shooting cans  in the Citadel. The whole "I'm Garrus Vikarian and this is my favorite spot on the Citadel" felt like a direct jab at my Shep in ME2, who was an advertising S-L-U-T for multiple shops on the Citadel.

Obiously all the Joker stuff was classic as well. I really feel like they brought the comedy hard in ME3, which is something I sorely missed in ME2.

Besides gems already mentioned, anytime I feel like someone's made my Shepard feel horribly awkward. You can tell because she offers NO response and pretends it never happened.

Jack: "Bite me, Garrus! Better yet, bite her. 'S probably how she likes it."

Javik: "You and the turian, you are joined?"
Shepard: "You could say that."
Javik: "I don't. Your pheremones do."
Shepard: *looks straight ahead and goes silent*

I don't think that was the exact conversation, but Javik's response to Shepard had me laughing.

Hahaha the bite me comment from Jack is definitely one of those "wtf" moments that left me laughing afterwards.
