After Anderson dies in a very tense, emotional scene...Hackett breaks in on the radio, and male Shep gives this "Jesus Christ...." intonation when he says "What do you need me to do now?"  Just something about that character choice, after such a tense scene, made for some tension-relieving laughter.

I like the subtle humor Javik tends to get into, the joking around with Wrex, the majority of the dialogue with Joker, and pretty much anything funny with Tali, especially when she gets drunk.  Heh, I'm getting too general though, aren't I?  So many great lines in this game.

I'll just go with the one I linked, and the drunk Tali dialogue.  I loved them.

I think after Cerberus' attack on Citadel, you can found Liara sitting in Apollo's Cafe inside Presidium Commons. If you persuade her to talk to her dad and you will keep visiting the bar, you'll eventually hear it. And before that, you should talk with Matriarch behind the bar (to hit a theme of Liara).
Though, I am not sure if it is accessible only after LotSB where you actually see the picture of Matriarch staring at Liara's picture and it is a chat starter in this case.

Javiks dry humor was hilarious all the way through. Garrus and Jokers alliance vs. turian jokes were pretty great too.

"Badassfully. Hey, man, that's my sister." Actually, the entire Blasto 6 awesomeness. If it was an actual movie, I would have gone to watch 6 times or even more. I also liked the "I am Garrus Vakarian and this is my favorite spot on the Citadel" line. Come to think of it, there were lots of hilarious moments in ME3, way more than in the previous games. However, they still can't cover that mess of an ending.

ME3 is full of hilarious jokes, usually linked to events in previous ME installments, but one which comes to my mind is:
Jack: "Oooh come on Shepard, everyone knows you can't dance."
* Shepard does that masterful dance performance again*

And Javik is a great source of nasty fun. I loved how he made everyone grumpy in a nonchalant way. All these "Oh, I remember when your kind was a group of sleazy barnacles. It's surprising you learned to walk." lines followed by the die-you-douche look, just a one big LOL.
It makes me think what Matriarch Aethyta would say him, haha.
