I'm a woman and almost always play as the female protagonist, about 98% of the time (100% for first run through a game).
In some games I will play a male character to get a look at the gender differences. However, I simply don't feel immersed in the character when attempting to play a male like I do when playing a female character where I can inject a bit of my own personality into the character during the game.

I'm male and started out with BroShep on ME1, standard vanilla version, romanced Liara and enjoyed the game.
Come ME2, I've heard people raving about FemShep, so I roll a new character -- and am blasted away by Jennifer Hale's performance. To me, it changed the whole dynamic of the story with a strong female character in the lead, saving the galaxy. (And a good "strong female character" at that who doesn't feel the need to prove herself to be as good as/better than a man.) That added a lot of fun for me.
As a bonus I learned about reach and flexibility with Garrus.
Somewhat immersion-breaking but highly amusing: Watching Femshep run around the Normandy in a black cocktail dress, especially when sits down and the camera peeks up her skirt. First time I saw that, I nearly fell out of my chair laughing.

I'm a guy, and so is my main Shepard. I'm transgender, and at the moment I spend every day effectively being forced into roleplaying as a woman (I don't read as convincingly male yet) so in a game with a custom character creator, playing a woman hits a little too close to home for me.
That said, Jennifer Hale does a great job, and I do have a female Shepard playthrough started, she'll just never be my main/personal 'headcanon' Shepard.

I'm male, my canon Shep is the default BroShep. I've started a second playthrough as FemShep though (1st ever!) to see how it all differs, if at all. Even early on I can say that Hale's VA is decent.

I'm female and play femshep, both for immersion sake and the fact it's just awesome to have a female player character who is actually taken seriously and not just there to give the guys something to ogle at, I like how it reminds me of the Alien series. I'll give manshep a go eventually but it'll take a while to get used to his voice acting (STEEEVEEE!!).

I'm a woman...well, it's complicated; I like my games with as many flavours as possible, so I roll manSheps and womanSheps as I feel like it. Their gender isn't that important to me; their personality and back story and my vision for their path through the game is what's most important.

Female/male Shepard. I prefer Meer's reading of the character; also, if I'm going to be looking at a character (usually from behind) for hours/days I want to enjoy the view.

Gender and sex are distinct concepts, and you cannot substitute the former for the latter just because sex is a "bad" word. If you unsure about the difference, I'd suggest buying a dictionary. And, yes, that also applies to Bioware writers.
