Why in the hell would anyone want to go back when we already know the end of the story. Some might make the argument by using a Halo Reach comparison, but Halo is different. They were already at war with the Covenant. Bungie left room for prequels with the way they told the story. The Mass Effect story is different. The Rachni wars are not interesting to say the least.
I played Mass Effect because of Commander Shepard and without him/ her, there is no Mass Effect in my opinion.

Noveria was probably my favorite place in the entire trilogy. I'd be down for seeing more about our psychological profiles though.
You could show part of Arcturus Station when Shepard is getting debriefed or leaving for shore leave. Then we could see see Akuze, Elysium or Torfan in more detail. I'd love to see a DLC like that. You could fit it into the narrative easy. Just say it was a dream Shepard had

The most exciting place I would like to see is a decent ending.
Once that's fixed, then I'd like to see Thessia, Palaven and a more complete Citadel (perhaps all of the zones that were opened up from the last two games).

The one thing I was expecting at the end of the third game, was some kind of understanding of the thousands of species which had been destroyed by the Reapers. That's where I want to go in the past, somewhere new but lost.
Maybe a mission with a long forgotten species which began the crucible (assuming it was a real event which resulted in an ending, and not the inconclusive crap we got instead). A hopeless battle for which the protagonist couldn't know the impact, but one that was of primary relevance thousands/millions of years in the future.

From humanity's history: Playing as one of the first humans on the Citadel after the First Contact War.
From galactic history: The Krogan Rebellions. It'd be interesting to play as a Turian during a time when there were no humans anywhere.

Standing right behind Spacekid, two minutes before Shepard arrives, while Spacekid is peering over a platform, down into a deep abyss.
Marauder Shields tried that but he slightly mis-timed his spacetime teleport and ended up on the wrong side of the Citadel conduit. true story.

Well, here is an idea of why the Illos conduit is no longer working: The conduit had to be an equivalent of a primary relay because it transported you more than half way across the galaxy (thousands of light years). Primary relays are not omni directional but have to be specifically linked with one partner. It is feasible to assume that when the reapers moved the citadel to earth (however they actually managed that) they displaced the citadel portal of the conduit so dramatically that they broke the connection. Just my two cents here so feel free to speculate

It's not active. In fact, it shuts down permanently if you wait around on Ilos too long; Saren and/or his geth sabotaged it to make sure that nobody could use it to follow him. (But he did so too late, and you were able to use it and continue chasing him.)
