Does the multiplayer missions affect the single player in negtative/postive way? If I screw up on multiplayer will it be bad for the singleplayer?

Nope. Multiplayer can never negatively impact singleplayer.
There are two ways it affects single player. Every promoted character gives you +75 military strength. Multiplayer also raises your Galactric Readiness. Galactic Readiness times military strength = EMS. If you have enough EMS, you get a very slight twist to the ending of the game. You probably know this, just saying it for completenes.
