After completing the game I started a new game with my existing ME2 character so that I could reach level 30, but I did not complete the game with the imported character. If I import this character to ME3 will the choices I made in the first play through transfer, or do I have to complete the game again with the imported character?

Depends what you mean by 'complete the game'. ME3 only accepts saves post-Suicide Mission. Your original game and your New Game Plus will be seen as two different characters post-Suicide Mission.

I finished the game, through the suicide mission, and then started another character. Do I have to finish the game again with the new character to be able to transfer?

Yes. Simply because ME3 only accepts post-Suicide Mission saves. If you haven't completed the Suicide Mission with your new character then that save will not import; that holds true even if you start a New Game Plus character.
