For me it has to be Luna from Lunar: Silver Star Story Complete. That game did such a good job of makeing you feel for Alex and want to save her.
I guess a close second would be Jack from Mass Effect. I loved everything about her.

But yeah over all I just don't like most "Pick a lover ones" Either they are stale, marry who you want just because (skyrim, Fable) or "lol say nice things and give me stuff" like pretty much any BW game.
Just is no substance, no story, no feelings. Most of those I only do for points, story or perks. I have no real motivation to do them. Story ones I find are more common for me in early BW titles, like getting my monk guy hooked up with the Empress in Jade Empire. Or getting my fem light side revan hooked up with Carth.
But over all I prefer the ones that are well wrote out and fixed.

Possibly my favorite of all time. Someone who starts out horrible/evil because they grew up in a world where you had to be Horrible and Evil to survive. But through the course of the relationship you show them another way. Learning to love something changes and nearly destroys her. Making unselfish decisions out of love for your character is a huge turning point.
2. Isabela (Dragon Age II)
Follows similar themes to Viconia, which is probably why I love the relationship. But does it with sexy Pirates. It was like Bioware put this relationship in just for me. Thanks Bioware!
3. Tali (Mass Effect Trilogy)
This one follows another romantic theme I love. Two people independently falling for each other but thinking the other one couldn't possibly feel the same way. When they figure it all out they get adorable together. I also like it because the attraction is more based on strength of personality than looks.
@spiderfan: I think SIE is one of my wife's all time favorites. She loved that scene.

1) Garrus Vakarian (Mass Effect series) - The best romance Bioware has made, hands down. All hail the bird man!

2) Sky (Jade Empire) - Just a fun guy to have around. And way better than Carth Onasi.

3) Leliana (Dragon Age: Origins) - My favorite female romance in a Bioware game. Such an adorable and badass woman.

Jaheira on the other hand was still is the best romance I've experienced in a Bioware game in my opinion, since there's so much time devoted to let it evolve in the course of the game. All the while keeping her basic characteristics.
TBH after Jaheira, I haven't really been bothered with the romance aspects. In Da:O it was just a whim of mine to try them out. Wasn't impressed. Leliana's one was sweet enough though, for what it was.

Yuna (Final Fantasy X)

Lulu (Final Fantasy X)

Leliana (Dragon Age Origins)
Ming Numara (Lost Odyssey)

Ashley Williams (Mass Effect 3)

Tifa Lockhart (Final Fantasy VII, pic is from Advent Children)

Anya (Gears of War 3)

Sheva Alomar (Resident Evil 5)

Oops I was only supposed to pick 1? I'm not scrapping all my hard work now.

Though the remark that she's "more comfortable with a firearm than a bra" seemed kind of tacky, given how comfortable she clearly was with her bras.

Love everything about him. In both games. Always.
And some more: Dawn Star (Jade Empire), Alistair (DAO), Anders (DAII), Fenris (DAII), Elliot (Fable III....finding him again...loved that quest...yeah, yeah, nothing much after that), Garrus (ME2&3), Kaidan (ME series), Wulffren (White Wolf of Lokken Mod for Morrowind), Chani (Dune 1. The really old one....):

You took all three of my answers
I loved the romances in BG2. What they lacked in graphical depiction, they more than made up for with build-up and dialogue.
I would also nominate: Bastila from KOTOR, because the good / evil paths that you could take with her were so pronounced. It gave more weight to your dialogue choices.

My favorites actually came from user-made Neverwinter Nights modules. Though I haven't played it for a long time, I do recall fancying Anera from the Shadowlord/Dreamcatcher/Demon series modules.
Have to agree with you on this one! *thinking of aDwR-mod*
Otherwise I liked Isabela (DA2) the most.

Liara was such a beautiful character in ME3. She was much better than her ME1 character, IMO. She was so sensitive and emotional in ME3. I just wanted to hug her...all the time but couldn't...
Tali, well, she's a woman of mystery. She wears that mask all the time and that's cute. Plus she has a adorable personality/voice. Who doesn't love Tali? Her romance scene in ME2 was beautiful. The woman was willing to take infectious disease and risk death just to make love to me. If that isn't commitment, I don't know what is. I refuse to watch the ME3 scene. I don't want to know what's under her mask, I like her the way she is. I'm not even sure if she reveals her face in ME3...but I skip the romance scene. I'd rather not have her image ruined.
Oh yea, Liara and Tali also never question my authority. Even when I'm an ass on some parts...they just go with it...

That's Trip, from the game Enslaved. Personally, I wouldn't call her a love interest, either.
