i Vary too much to consider one as best.

my most Origonal one is my Falcon Human Soldier.

Uses both the stun on the Falcon and the stun on Concussive shot used at the same time to stunlock anything. its the single best weapon class synergy i have beyond the player controlled Geth Hunter (Geth Infiltrator with a Geth Plasma Shotgun)

the only problem is that the damage on it is extremely low... can walk right up to most enemys spamming rounds tho.. the stunlock is so constant that most enemys cant react, but it takes a dissappointing number of shots to have ANY impact on a boss.

i also reecently found that having the Krysae on the Krogan Soldier is a great Crowd Controller in the same fasion as my Human Falconieer. plays compleatly differantly tho.. and is best at range.

Most effective: Geth Engineer w/ Geth Plasma Shotgun (no second gun - Thermal Clips mod instead). Turret specced fully healing is the key to effectiveness (can't win if your squadmates die). Anytime I'm not doing turret maintenance, I use Overload + the GPS to do some damage. I can still do surprisingly decent damage while keeping people's shields up, but what really makes this build effective is the other three people on the team who can now do riskier things and get out of worse situations.

(If you want extra detail: I skip Hunter Mode entirely, which makes me and my turret spamming powers rather non-squishy and lets me max everything else. Full health/shields on Fitness, weapon damage on class skill, and Overload with Chain/Recharge/Shield Damage.)

I don't think I'd call it the most entertaining, no, but it depends on mood. Sometimes shooting turrets around to heal my teammates is what I find most entertaining that day. Other days Phoenix Vanguard is really fun, and I love to play a Turian Soldier and spam Marksman when I just need some more dakka in my day.

After seeing some of the replies about defining the question, I should amend my original statement.

My most effective build is, in fact, any infiltrator with a Valiant. My most fun builds are almost anything else.

In terms of effectiveness, nothing beats a Claymore GI, no explanation needed I guess.

In terms of pure entertainment (but by no means bad):
  • Drell Adept + Talon, simply because I love those instant BEs, Talon is a beast at close range.
  • Asari Vanguard + Graal. Charge Graal, charge yourself (I get surprisingly many headshots out of that combo), charge Graal again to finish anything that is still alive. Oh and then there are those shiny 'nades.
  • Geth Infiltrator + Scorpion. Added this one because of the confusion it causes on pugs. "Why is everything exploding?"
I'd probably add the male Quarian Engineer to that list, but unfortunately I have maxed both Phoenix Dudes, the Vorcha Sentinel, only lights missing on the Vorcha Soldier but am still missing both male Quarians. RNG is such a troll. :o

I play all classes and each class has its own weapon combo that I use for "serious" play but I also like to use odd ball choices and experiment with all weapons. I do "serious" play on Gold and Silver. I really enjoy, however, playing oddball stuff on Bronze in a more relaxed atmosphere. What I'd like to see is more more targets in Bronze. NOT harder, not more tense just more targets. Sometimes there is no one left to shoot.

Definitely nope.

Sal-Inf/Geth-Inf ends up being boring for me. You're just staring down into a barrel, shooting fish. Though I do like rolling with my Hum-Inf with the Reegar. (My god, the damage, and it's only a I.)

That would be Vanguard

There's something about being able to spot an enemy from across the map, shout "Hi!", Biotic Charge to him, shotgun him in the face, then yell "Bye!" as you Biotic Charge his friend. Sadly, Novaguard is easy mode and Drellguard is like going to a S&M Club without a safety word and Kroguard's recharge time on powers is astronomical. Coupled with the fact that you play Vanguard YOU MUST HOST? It's an experience I can't readily enjoy.

No, there is a difference. One is subjective, and the other objective. Opinions =/= Facts. You can easily find out the most effective class using in-game data, but not what individuals think is the best one.

Most Effective: Geth Infiltrator (build)

Most Fun: Human Vanguard (build)

Was messing around with Human Soldier builds and this one turned out to be fun on Silver, but I doubt it would be any good on Gold.

WTF Human Soldier

The weapon combo would probably get me kicked on Gold anyway :P The idea is to stagger enemies to close in and melee the survivors.

with a Falcon, Claymore, Saber, or Harrier

A glass cannon but so much fun to play

Phoenix Vanguard

Charge: Radius, Power, Barrier
Smash: Electrical, Damage, Armor
Lash: Detonate, Damage, Shield
Phoenix Training: Capacity, Power
Fitness at 3
with a Disciple and a Phalanx or a Carnifex

Smashing everything at close range, Lashing everything at long range, and Charging when Shields are low.

Asari Vanguard
Charge: Radius, Power, Barrier
No Stasis
Lift Grenade: Damage, Max, Damage
Asari Adept: Capacity, Power, Weapon Damage
Fitness: Durability, Shield, Fitness Expert
with a Disciple

Charge, shoot, Grenade, back dash------> repeat.

Asari Justicar

Biotic Sphere: Radius, Weakness, Warp
Reave: Radius, Damage, Duration
No pull
Asari Justicar: Capacity, Headshots, Assault Rifles
Fitness: Durability, Shield, Fitness Expert
with a Cerberus Harrier or a Falcon

Reaveing, Biotic Explosions, and Shooting kills everything.

Anything that depends on aim... I can do it, but I will hardly be the exemplar of how it's done.

Mind you, even with that, I'd probably still name an Infiltrator as my second most effective class. They certainly are good.

My most effective is:

Geth Infiltrator specced 4/6/6/6/4. Claymore X(Smart Choke + Extended Barrel). Shotgun Rail Amp V gear with Shotgun Rail Amp III, Warp Ammo, and Power Amplifier Module.

However, I never use it unless we're trying to do a speed run as I don't find it as fun as the other builds I use when I'm just playing normally. I have quite a bit of fun playing as my Human Soldier, using the weapons that are considered not as good around here.

I also love anything to do with Quarians =P

I will fully admit, my skill as an infiltrator leaves a lot to be desired, so I could not be honest if I were to toss my hat in with any who place that as their go to class in that category.

My human Sentinel with Phalanx X with EB/AP mods. It's not the most effective, but it's the one I have the most fun with - especially with any other biotic class to work with. Either detonating their explosions, or having them do the same for me, is just a ton of fun. :)

Second favorite is going to be Turian Sentinel with Vindicator (AP and Stability), and third is human Engineer with Mattock (AP and Scope). The powers I use for those two change depending on what I feel like playing, but they're always fun. :)

Most Effective:

I haven't played one in a while, but Geth Infiltrator with a big gun and some melee spec was shockingly powerful. It wasn't as much fun to play, in part because Hunter mode is annoying after one match.

My favorite (Gold) classes currently are:

Drellguard 6/0/6/6/6 and he's a ton of fun unless there are other grenade users on the team. I my respec for pull soon.

Krentinel 6/0/6/6/6 with Falcon and Claymore. Effective on a good team. Always avoid geth for reasons I'm sure you know about already: stunlock, 'staggered' hunters, rocket trooper who shoot two rockets out of the back of their head.

Kroldier: 0/6/6/6/6. This guy is a blast against Reapers. Fire explosions everywhere. Do I miss fortification? Not, one, bit. (And again, geth suck.)

I was going to suggest playing console as the bullet magnetism helps, but you're prolly on PC.

Other than that, it's not hard to play. Spec Sal-Inf for power damage and just do a rotation from cover: Cloak, ED, bodyshot with Widow and you'll kill most anything that isn't a miniboss.

What does sniping have to do with it?  Use a shotgun.  Use an assault rifle.  Use the Hurricane.  It doesn't really matter.

No matter what anybody says, the Infi is golden and should never change. I love hearing kiddies whine about how cheap my class is, only to revive and carry them every wave, and then they kick me because I had them WIN.

Right now I having the most fun the Quarian Male Engineer with a Flacon X. All that AOE damage just makes me smile. Adding either Cryo, Disruptor, or Incendiary ammunition just adds to the fun. Really good for blowing up Phantoms, either singly or massed.

Im most effective with the GI with a claymore. Despite being the setup Im best with I dont like using it very much. Makes things easy and boring a lot of the time especially when playing with a good team.

The class I enjoy playing the most at the moment is Human Engineer.

For effectiveness: 
Depends on difficulty level. On Bronze (which is mostly about who finds the enemy first) a Human or Krogan Vanguard will always rule (since a Vanguard will always get there first) :D
On Silver and Bronze, when it becomes too risky to use Biotic Charge as a means of transport, Infiltrators take the top because they just deal insane amounts of damage and don't get shot at.
Asari Adepts can be extremely powerful provided there is nobody cancelling their biotic explosions with Overload or something.

For fun: 
Personally, I have the most fun switching classes after each match and going through the entire spectrum in a handful of games. Any class will get boring if you use it too long, so I mix them up as much as possible. I tend to stay away from the more powerful builds because I feel like a jerk using them, so I tend to only use the Kroguard solo and equip all my Infiltrators with the Javelin (arguably the most useless of all Sniper Rifles since Shield Gate) to take away some of their advantage.
