I'm starting a new playthrough (wanted to continue last playthrough but wanna try Adept) and I have completely no idea of the choices you can made (colonist/earthborn/spacer and etc.) because I have never played ME1 and ME2
so what is the impact in ME3 if I choose:
Colonist/Earthborn/Spacer?
Sole Survivor/War Hero/Ruthless?
Suggestions on a new playthrough

You played ME3 before either of the other two? Heresy.
And as far as I know, there isn't a huge impact depending on which origin story you choose.

Will these two appear only if playing with a ME2 imported character?

Jack dies in ME2 and so does Kelly if you don't import

Yes. If you didn't have a save game to import into ME3, the game plays a little differently. Kelly and Jack being omitted are a part of that difference.
