I generally play with my friends. It feels like I can't trust random players on higher difficulties.
Do you prefer random groups or regulars?

Randoms. ALWAYS.

I enjoy playing with friends and randoms both. Most of my Origin F-list is people I played with in PUGs. I usually have more fun with friends though, since PUGs tend to put me with laggy and long-distance hosts.. and random farmers who seriously aren't ready for higher difficulties.

I only have one friend really that I can play ME3 with so whenever we see each other online we join up, just 2 of us. But I play plenty without him and I don't mind randoms at all. More often than not I find a cooperative group, no farmers.

Mood dependent. Do I feel like talking?

I play almost exclusively with friends. If we have an open spot in the lobby we'll usually let a random join. The game is just more fun with people to talk to.

Always randoms.
I play an MMO in a raiding group, so this is my no-pressure stress relief game.

99% PUGS. 1% friendlies. I like to be amused... and maybe a bit masochistic.

I prefer friends because games work better when it's people you know and you can play to each other's strengths and weaknesses. I'm mostly forced to go randoms when I play because all my friends have moved on and most of my FL will be playing Borderlands 2 tomorrow anyway.

Well, generally I prefer to play with people on my friends list, mostly so I don't end up thrown into Firebase White Geth...
I would like both to be viable options though, I like playing with randoms because it shakes things up. Sometimes you'll end up in an awful group but then sometimes you'll find some really good players and stick with them for a while, and maybe send a friend request.
Pretty much this.
The majority of people I play ME with are from the BSN forums or groups so are well up on the mechanics of the game making most matches a doddle.
I enjoy playing with some of these people so much that I have actually, of late, found myself in the slightly shameful situation of playing GoW3 just to get a few games in with them. Previously an avowed multiplayer-avoider, I have been ruined by Mass Effect.
Given the choice every time I'll play with people from my friends list- I play the game for fun and it's infinitely more so when you're laughing and chatting, whether about game mechanics or random nonsense- but if people aren't on/don't want to play I'll happily do a few randoms.
It's much more of a challenge playing with people whose style of play you don't know, and who don't communicate- but just how much of a challenge I want depends greatly on my mood. Today, for example, was very frustrating- playing revivebot/objectivebot for 10 rounds and then having people step over your corpse is a bit of a jaw clencher. Other days I love running around being left to do everything.
On the whole those rarer occasions where you'll get a random Team of Awesome where people throw turrets/bubbles/decoys up to protect you while doing objectives, or switch their character on seeing yours in the lobby to synergise better, or run as a team (using missiles) to revive fallen team mates make the frustration (mostly) worth it.

I enjoy playing with friends. Usually only have a group of three who want to play so we always have a random in the game just to mix things up. Randoms can be fun.

I actually enjoy playing with a random team, it is more challenging. When trying out strategies or quickly farming platinum for credits I play with friends.

Regulars, that way if I'm trying to learn a new class and having a game that turns into a nightmare, no ones going to yell at me. Well they might, but if I don't hear it, it doesn't happen right?
If playing with randoms, I'll play tanky characters where I know I can hold me own.

If I'm on a mission, friends. I'm always on a mission. Randoms are too unreliable. Randoms weren't always so bad. I used to play with randoms most of the time some months ago.

I like both, but...
...if I play with PUGs, I have no way of knowing whether I'll have a good connection or not, and the game experience can be utterly horrible. Quitting means losing equipment, so I have to decide between running without equipment (which seriously limits some of my favourite setups) or risking wasting it on a terrible game.
...if I play with friends, there are frequent and consistent problems connecting to each others' games via Origin, and some of them (hi xtorma) get disconnected from the EA server repeatedly without warning despite having a rock solid connection to the other players.

*sighs* Poor puppies.
I promise you will be informed as soon as there is something to announce. For us, that means, if there is anything coming, it has been approved on all fronts.
Cheers

I'm all for randoms since I hate having to wait for people to get online so that I can play some matches. It's a way for me to try and break the monotomy of playing the same 8 maps over and over again. Plus, I don't have to worry about "Oh dude, I can't play tonight because I have a life" or "Hey, I can only be online like 8 hours after you are". I don't like having to adjust my playing schedule to accomodate others. I play when I feel like playing. If I don't want to play the game for 2 months, so be it.

I tend to play all randoms. I've only got a handful of friends - most picked up playing MP, but they're rarely - if - ever online when I'm online.
