Don't expect any carry over characters since they have become, unfortunately, baggage. Heard someone whisper "Shepard is considered a legend," so the timing will probably be not too long after ME3... maybe a team made up of some of the races who survived the battle of Earth. Also, if you picked any ending other then "destruction" I suspect you'll be disappointed.
Of course, if they wanted to clone Shepard they could. I'm sure he left DNA all over the Normandy, though I doubt that will happen until ME7.

If one would be overly cynical and believe they will bork the next one... like me... ME5 could be a good guess too.

If they can do MENext with no references to anyone or any events from the original trilogy, it'll be a better game for it.

▲▲ I would be disappointed if there was no mention of characters or lore of the prior trilogy and how it effected the new game. It's not Final Fantasy, it's Mass Effect.
Well, the GM of BioWare, Aaryn Flynn, said there would be nods and references to trilogy in the next Mass Effect. Interpret that how you will

My money says it's not going to be called ME - A Clean Slate.
Also heard there's supposed to be one main villain - maybe one of the Merc groups taking over a couple of systems.

Exactly my thought on the matter.
Too many game devs these days try to get the desired emotional reaction by deploying downers. It's a cheap way to do it. As if a positive spin or hell, even bittersweet won't accomplish that. It's even worse if they are supposed to mean something if you read between the lines but they actually don't.

What made Mass Effect special to me was the profound sense of wonder and amazement that drew me in when first landing on Eden Prime. When it grabbed my attention and made me think about other possible life in the galaxy, was when I was hooked.
I love the trilogy as a whole, that's just brilliant story telling and excellent execution. Talking about it years later and I still have the same enthusiasm as I first did.
I think it's a common idea about exploration for the next Mass Effect game. I love this idea but I hope it carries that same wonder and amazement as it first did with me. I want to be drawn in to strange missions that lead me to a previously unknown technology or even species (perhaps a first contact party with great consequence if handled renegade/paragon).
Another idea would be that during the first trilogy, I often played campaigns either as a biotic or a soldier (these being my favourites). I'd like there to be some story elements whereas your decision on that actually contributes to a story plot and individual discovery, make it more personal.
Other than that, I'm very grateful for these brilliant games, they actually shaped my own interests such as astronomy and that in turn shaped my whole outlook on life and Bioware creating Mass Effect was a great influence on that.

P.S I think post M.E 3 events would give greater flexibility in story telling, as much as I'd love to see what action could be drawn up from The First Contact War, we all know how that ended if you listened to the codex.

First of all port saves from ME3.I want to see where all my team went after the war.
Second:player must be able to choose his race to play and each race will have a start bonus.
For example:asari will have bonus in using biotics.
Salarians because of their fast lifespan will have kind of slowlotion.Ability to slow time during the fight.
Turians will have... well something else.
The world state depends on the player choices.If saves were not ported then its default.
Will bring more ideas.
The exploration from ME1 must be returned and expanded.
Player must be treated by NPC's depending on his or her race and gender.
(Some people dont like turians,some dont like humans e.t.c)
Also party members must have great variety(different races) and thie own opinions on
the situation.And ofcourse romances.Also player must be able to influence party members to
change their opinions more to player likings.For exampe turn a pacifict into a cold blooded killer.
Weapon and gear:fully customisible and upgradeble.More kinds of upgrades and weapons.
Non-lethal weaponry for hostage missons.(I remembered biotics mission from ME1)
Player ship:space combat and upgrades must be implemented.
Will bring more ideas.

I would love to see that we get to witness the after affects with the war against the reapers a of decades after the end of ME3 where planets are ravaged from it, governments struggle to regain control of their territories, huge graveyards from battles in space and on planets with reapers. we also get to see how earth has been altered from the war where some places have benefitted from the technological breakthroughs with the reaper technology, while others are turned into huge slums consisting of humans and other aliens. the new threat could be a barbaric alien empire, who have destroyed dozens of planets and caused the extinction of several species, comes to conquer the weakened races and explain why they only destroyed the advanced races only. the way it could set the story up is allow us to make a completely customizable character and background like Guild Wars 2 and this also would go nicely with co-op and depending what race we chose we would fight for them against this threat and it also would allow us to see the perspective what some races think of the others like you could play as a turian who still feels some resentment to humans form the first contact or a krogan who is only fighting for the money. multiplayer could be almost like a gang war where you make your own character and play it like SW battlefront style.
this my opinion so feel free what you think of this and comment

after they (who ever it was?) brought back shepard, the idea of 'the dead' left over from the reaper war (depending largely on what choices were taken) seems like a bit of weird science?
I wonder what kind of elevated music they'll play on space elevators?

I completely understand the attachment to all the squadmates and friends of Shepard, but I never quite understood why so many are gonna miss Shepard. In ME1 and ME2 he's merely the player avatar. The only characterization he has is the history with the aliens and human friends that you give him. In ME3 the writers decided to make him a real character but he pretty much became a Mary Sue. Everyone is like "I don't know how you can do it, you are so amazing Shepard" and then because he fails one mission (thessia) he starts crying almost. What a self-centered dick, man.
