There was so many posts so I do not know if there any ideas related to what I am about suggest.
1. I hope writers will be loyal to their own thoughts of the story.
2. Something like moving with protagonist's love interest or best friend to see how they view protagonist's situations personally. Like when the hero him/herself
is temporarily captured or sick(coma?).
3. Some serious survival missions would be cool. Just to see your party being badass.
Well that's what I have I hope to see. But number 1 is the most important.

That we want the writers to get involved with what they want and not just do what we want is kind of a given to me. I've loved the games so far, with and without controversies, they made me feel fuzzy, angry, enamoured, sad, all in all: a little more alive. So of course, of course I support the writers and makers of Mass Effect to explore where they want to go.
What I personally would want to have more of - besides a picture of Shepard and their crewmates in a bar in the Caribbean, reunited and happily drinking away, with Tali and Garrus making out in the back and my lil indian femshep with her love-interest (Kaidan) + Jeff with a reproduction of EDI and an ice statue of Mordin - I am drifting. Making a list should help my purpose:
1) Besides the whole exploring a completely new area, planets, etc. (in which it would be nice to not have the kinda boring minigame-ish thing of scanning 100 times, which is fun the first few times but gets old quickly) I of course support deep characters with their stories. I really loved and appreciated the Citadel DLC, because you really understood how not only the people on your crew were your friends, they were close to each other, they were a team, and they backed each other as much as Shepard. That was an amazing feeling, seeing and exploring these personalities, and creating friendships with and between them. The Mass Effect characters are interesting because they're smart, funny, different, weird, awkward and real. Make us love our new companions as much as we loved our last ones, and I will gladly bow to you.
2) Apart from that, bigger philosophical conflicts such as Geth vs. Quarians or Genophage: Necessary or Evil? (which I consider being one of the most wonderful achievements of the makers of ME, the gray moral areas of any conflict like these, the discussions with your crew members about these conflicts, later discussions outside the game with friends and the community etc.) are high on the list of what I'd love to see more of. It's difficult to create conflicts like these without people going "oh, but this is like this real life conflict", but the ME team did it. And I think this was beautifully solved.
3) Krogans and Salarians. Who didn't love Wrex or Grunt, and who can't agree with "I love you, Grunt." - "Heh, heh, heh." And who didn't love the Salarians much more after we got to know Mordin? There's just something about these amphibians ("They used to eat flies.") that you cannot shake. Whenever and wherever this new game takes place, some winks and reminders, new-comers of these races would be heart-warming.
So all in all: Well-written characters (as always), that are to be deeply befriended, interesting gray moral areas to be discussed and maybe even influenced, and charming races like "the fighter-race, that's still funny and loveable" or "nervous-but-crazy-smart-and-funny-amphibians". On a sidenote: I genuinely think it's cooler that these characters, which could have been made as one quirky/brutal/... human, are cooler being made as a whole race.
So stay awesome, go with your gut, and give us a good, heart-warming game with a happy ending.

It just occurred to me rewatching the teaser and doing Bring Down the Sky again:
A radio station, like in Fallout.
Maybe include easter eggs like Miracle of Sound's Mass Effect inspired songs or soundtracks from the previous games via it.
I mean, M4 part 2 was an awesome closer to Mass Effect and just awesome in and of itself, and if you haven't listened to 'Reignite,' 'Take it Back' and 'Normandy,' why are you here? Go give them a listen, then come back.
Not sure how hard it'd be to code, but at the same time, it'll give us something to listen to besides engine noises while we're driving the Mako around on side-mission planets.

Idk about you guys...... but you shouldn't really just throw away Shepard that easily........
How bout this....... Mass Effect trilogy is kind of similar of DAO [ yes i kinda noticed that, uniting nations, against an unstoppable force, you can either sacrifice yourself or be alive, strong race that isnt allied with any other races, darkspawn/reapers uses their own versions of other races, ]
Mass Effect 4 can either let you play as shepard if he is alive or play as a new N7 commander or a new race/character.........
Fair for all ............. aside from massive amount of work from you guys ......... i think it would mend the effects of the ending of Mass Effect 3........ You know, just like Dragon Age Awakening ....... [ also satisfies people who wants a new start or wants to continue ]
This is from a full-time player of the trilogy .... and i dont think just throwing away the main reason why the Mass Effect trilogy is even recognized that much is good ........ Also some cameos would be good if from a different galaxy but if it were the same galaxy then ok........... Don't be lazy bioware ...... All of us got heavily attached to shepard ........ And if someone feels disappointing about it then it can go spread around like a plague.... It says a new mass effect game so this isn't heavily directed to Mass Effect Andromeda sooo yeah
But then ......
- New powers [ the more the merrier ]
- More fluid combat [ i like to move around and not feel stiff in the heat of combat ]
- Bring back the overheating mechanic from ME1? [ it was sooo unique and didnt feel like just some classic shooter ]
- More dialogue between companions [ and like i said, the more the merrier ]
- More weapons and armor and mods probably
And if you guys are going for a new galaxy then this
- Make more memorable and awesome characters than Garrus and Tali [ JK , they're the best NOONE could replace them ]

I just have finished the ME trilogy 3 days ago. I know It's too late when compare with a lot of guys on this forum.
IMO, ME trilogy is probably one of the best RPG franchise created by far. It has a lot of unique elements: Dialogue wheel choice, Paragon/Renegade system, Combat system and of course, the unforgettable journey of a hero name Shepard !!!
So in new ME, do not throw those elements away. You do not need to be a free game like Skyrim, your worlds does not need to be a world of "evil-lesser evil " like The Witcher, just be the Mass Effect we enjoyed: Paragon/Renegade choice, 6 classes ... and finally, Shepard and The Normandy's crew.
Please do not throw Shepard and his crew that easily. We all want to know what will happen with those blue childrens of Liara, Tali's house on Rannoch, The Krogan after The Genophage cured ... You guys created a lot of unforgettable characters, and now you guys just tell us to forget them, I think It's not fair. I also know It's hard to you guys Bioware to create a whole new story can cover all endings possible in ME3. I have an idea, someone on ME universe discover a way to travel back time, but that travel is a one way trip, It can use only 1 time because It need a very rare element name "abcxyz". And also because of that element is too rare, It will allow us travel back a little few years, not unlimited years. The rest of the story is yours. Just an idea, It's a very popular motive, but I believe It will works !!!
One last thing, I think this depend on EA. I know DLC now It's become a must have features in a lot of games, I do not hate them, but please made only story DLC require money. And make your DRM more friendly, do not make us jealous with those guys who pirated your games, when they can skip those nonsense feature checking DRM in-game, 24/24 online needed ...

MMO vs RPG - please, do not focus on MMO because, IDK, i is popular. I mean, we did just fine without it, it´s NOT what makes BW a BW. If you gotta do it, that´s okay - but make it not on the expense of SP, and make it perfect. No pressre.
Characters - are what makes us involved, be it our character or companions, allies or foes, those stnd-byers who don´t do anything in particular, but makes the world real. Please, do not make interesting characters for the sake of it. Make interesting characters because people are interesting. Make us love them, hate them, love/hate them, make them surprise us by loyalty or betrayl. No need for touchy-feely BG, if it´s notin place.
Races - d not make HUMAN ALIENS! Make Trian value honor above all, make it a plot, think through each races psychology.
Fun - make us and the cgaracters have fun, but make it optional! The opportunity for shore leve, minigmes with the crew, gettin involved in what THEY are doin, perhaps studing with a scientist or hlpin a doc for NO xp.
Planets - see Races. No Earth-like bullshit unlss it is an exoplanet, thank you! Think of other lfeforms than a simple animal/plnt/fnghi scheme.
Ships - make us improve them, customize them both ext/int, let us by, let us have options for that, maybe make it an optional quest, make us fight for them with krgan!
