1. Play as more then one race.
2. More locations to return too, other then the Citadel every time. (illium, Omege maybe even Earth or other Homeworlds?)
3. New designed Weapons, because I dont know about you, but some of the weapons that have been through all the games are starting to look dull, time for something new i think!
4. Character Returns! I want to be able to meet up with Tali, Garrus, Liara and the others again!
5. Bigger squads. Taking two Teammates into battle is fun and all, but sometimes the dialoug between the main character and even the two NPC's is a little... Bad (Not in all places, and a small few.) So maybe 3 squad members to take with you will be intresting and a good variation.
6. More Animation. I have gotton a little annoyed at times just seeing the same head turning, hand waving and leaning animation so many times over the games.
Not everything I can come up with right now, but In my opinion, I would love to see the game based off of the Control or Sythesis Endings. Also, a character at the end says "Tell me another story about the Shepard." Meaning that maybe... Just maybe, Shepards Tale didnt end after the events of ME3. I can think of ways that he/she could return, with all that new technology. But it may involve a little Q.Qing. We will never be able to get a story which everyone is happy with, but we do have a character which everyone loves, because we shaped him/her in the way we wished. I will most likley make another post soon about my ideas storyline wise, but the list above is what I think that should be focused on.

Real consequences. I want a journey. Not a race.
The game consistently changed (or was from the beginning) into the following - hear part of plot - travel to a new destination, kill a bunch of baddies, discover more plot.
ME3 felt like "do this and do that and get through the content ASAP so that you could discover how it all ends".
Emphasize exploration and detective work. Allow players to discover new perspective in missions which can change the players opinion and choice. Need to kill a baddie? Sure, send us to his lair. But if we find that the baddie is doing "greater" good stuff - we will have conflicting opinions. Some will kill him cos what he does is pure evil. Some of us will sympathize. MORE OF THIS STUFF.
FAR LESS linearity. I BEG YOU. The story in the trilogy is "acceptable" of market standards but it is drying out fast and is no value of money. You only disappoint your already decreasing fanbase.
The choices are bland. There were a few dramatic moments where you decided the fate of very personal characters, but it all narrowed down to rails in the end. Rails. Very long, rails, right from ME1.
And the combat system. Less of "here, have half a clip of futuristic grain size bullets traveling at 10 mach and don't die". Emphasize on positioning and making advantage of the environment - not stacking multipliers in an attempt to break a threshold and kill stuff faster... More dynamic combat and less long, winding fights. Stealth, DX:HR style. Allow to avoid combat if player chooses so through redirecting paths and so on.
If you plan on milking the franchise - drop it. YOU will be disappointed.

Keep the game play the same just different prolouge missions that lead into the same main story like if you were an asari your prolouge will start on the planet thessia
And your choice from me3 will brought up in the story as a vi on the citidal

- Shepard deserve to go back, until his story will be honorable ended. This ending is just horrible for his story (satisfy, but still horrible).

New playable races... (all is irony and silly)
- Commander Krogan will truly awsome for example:
"The Child: Commander Krogan, can you help me to find my parrens?"
"Commander Krogan -> paragon option" I AM KROOOOGAN, no any child will ask me to help, it offends me!
"Commander Krogan -> renegade option" I AM MASTEEER KROGAN CHIEF and I ate your parrens to dinner!
- Commander Salarian will solve all your problems and make trilions more problems!
- Commander Vorcha is excelent speaker!
- Commander Turian... "Hmm, I'm turian and your are just a human... yes, maybe your race saved the galaxy but still U'RE HUMAN (and I'm rasist)!"
But maybe Asari and Humans can win this challenge, but everyone knows... that Hawke guy from DA2 is boring guy!
So, if you still want playable races, characters, play multiplayer or make your own MMO on unreal engine with your boring characters!
For singleplayer this can't work!

please hold on to the original thinking and style when designing or re-designing.
develop already known races and races' cultures.
incorporate things already seen in ME-triology with things that would be natural in the mass effect universe.
so whatever is made, make it in a mass effect way.

As for the style of storytelling, I want a return to the openness of ME1. In the second two games, the typical mission has you kill the baddies, and make a decision at the END of the mission which may or may not *cough* effect how future events unfolded. But in the first game, there were often alternatives to getting to the end of a mission. Noveria is a prime example: there were four or five ways to get the pass card to get to Peak-15; once you were there, there was three or four ways to find Benezia; and to top it all off, you get to choose whether to kill the Rachni queen. The point I'm getting at is the latter two games diverged at the end of the journey, but the first game diverged on the journey itself, well before it's conclusion. This is what made ME1 so replayable for me, the experience felt fundamentally different each time. In the second two games, I was almost always left killing the same people in the same hallways.
Similarly, return to variable mission orders. I know the second two games had technical limits being on two discs, but I've got to say I really enjoy being able to do the different major missions in a different order. The two-disc limit shouldn't be a problem since I'm assuming the next game launches next-gen.
Summarily, what I want is the tight gameplay and graphics found in ME2 & 3, but the openness, the mystery, and the "tangible biggness" (to steal Yahtzee's description) that the first Mass Effect had in spades.

Every decision the protagonist makes has an effect on the loyalty of individual squadmembers.
For instance, saving the rachni queen in me1 would have given Wrex -100 loyalty, Ashley -50, Liara +25...
At a certain pointcount disloyal members will start to become rude, stop talking alltogether, or even leave.
Romances can only happen with members with for instance +250 loyalty points.
This would make the relationhips a bit more interesting and add replay value I think.

1. a protagonist whose gender and race players can customize
2. A origin story arc much like how DA:O had
3. A story set far into the future like 400 years or so, I want this so we can get some distance and
have less overlap from the first trilogy and so gamers feel they can have a fresh start with a new trilogy that has its own problems and is not just dealing with post reaper clean up.
4. Maybe base the premise around a galactic civil war? Just a thought...
5. Would really love to have more customizable space ship options, like colors, emblems and furniture sets to make the ship feel like my own.
6. Side Missions that are not just courier jobs, bring back N7 missions I loved those!
7. Shepard was always sort of a brick, he really had no substance as a character, A new character should have more conflict attached to them and character substance in them, I feel like this was starting to be explored in ME3 but was not fully realized.
What I don't t want:
1. Reapers-been there done that
2.Prequels-I just dislike prequels.
3. Courier based missions, like the side missions in ME3 i guess you could call them fetch quests
but whatever i still hate them...
4. War assets- i didn't feel as if it was anything more than a statistic i had to manage, in no way did it actually feel like I was building an army don't really know why this feature was included i felt it
was unnecessary.
5. Revisiting old characters- don't get me wrong I like garrus, but i don't want to meet him again as a new character, don't waste time rehashing old characters bring me new ones that i can get to know, just make sure they can live up to the awesomeness that is Garrus Vakarian..just sayin
6. Endings-make them less about interpretation and more about closure.
7. Multiplayer-Multiplayer is nice but it should never take center stage over single player, i doubt it would happen but in any case I just have to say i don't want mass effect turning into a multiplayer centric game, its fun but the single player has always been the main draw for me thats why i buy mass effect.
