There is much to explore in the Mass Effect universe. Shepard's story was great, but it has been told, and because of the scope of that story, anything post-Shepard would be very challenging to write. It would likely be best to leave Shepard as is/will be (post-DLC).
Many of the historic events in the galaxy (First Contact War, Rachni Wars, Krogan Rebellions etc.) would provide a great setting for creating conflict, the driving force of any story, without requiring the game to be a 'prequel' per se. Another option, and probably the more interesting one, is a story that parallels Shepard's leading up to and during the events of ME1-3. This could allow for some horizontal continuity if save games were imported (for instance hearing a report of Sovereign's attack and the fate of the council), something that would validate a player's prior experiences in the ME universe.
As far as game ideas, there are many options, but I think playing a non-human Spectre in a shadowy 'intrigue and espionage' setting would allow for deeper exploration of the universe, and would allow for TONS of interesting plot lines. It could be lots of fun to both write and to play. For example, for a spectre who answers to the council, even just the repercussions of Shepard's decision regarding the Destiny Ascension would be fascinating to explore.
That's my two cents. There is much to explore in the ME universe beyond Shepard.
This needs to happen.
Agreed, this is good stuff. I'd add that BW should try to get away from the "Reapers or Arch Demon or other external threat that all must unite against or die" storyline. BW has the writing skill to explore less black & white issues and force the player to make more shades of grey decisions. Inter-species conflict / espionage seems like a way to do that.
Though I have yet to play it, Dishonored seems like an interesting example to look at, as it allows the player to accomplish missions in many ways, from killing everyone including civilians to complete non-lethality, and those decisions go on to have direct impact on the game's conclusion. That model could be adapted for spectre play.
And to add my own lesson learned quibble from ME1-3, I'd say if there's going to be another multi-game storyline, map the storyline arc out at the start and stick to the plan so there are no jarring changes of tone or theme or direction from start to finish.

If not for all the myriad implications of the two previous installments, ME3 might have been a decent, if forgettable (due to the lack of buildup and brilliant introduction from ME1) standalone game. It was almost a brilliant final chapter, save for the final scenes which discarded all the implications and foreshadowing previous to them, not to mention the vital themes of the series and its focus on Shepard (which was abandoned for some kind of lofty pretentious 'higher statement' which was derivative of other science fiction more than being profound as intended).
It's not that we're not capable of getting that. It's that it wasn't the story we were being told from the beginning. That was the problem. Consistency is important in a fiction series, and that includes games when their story is told with such a level of detail.

I think that an area of interest that could be explored is the vast power vacuum that would exist after the war. All the races poured all their assets into building that floating onion wand, The Crucible. With the such resources spent, criminal elements, interest groups, and many others could move in for the kill. Alongside that, the utilization of highly advanced Reaper tech would be a temptation for all. It has already been proven that the races hoard whatever Prothean tech they can to get an edge. What do you think they are going to do with giant Reaper hulls strewn about?
Another thing: Dark Space. The Reapers had been filling that border between separate galaxies for a very, very long time. Isn't it a little curious what could have resided beyond? Perhaps the Reapers were created in this galaxy, and being aware that they simply could not regulate ALL forms of life, the isolated this one, maintaining it as they saw fit. They could have known all along about bordering alien life, keeping it out and our side in. Imagine the possibilities of exploring that universe.

Republic Commando-styled gameplay and squad control. Free roaming in between missions to bars and other places. Chase down leads, beat up people and generally act like the goddamn turian batman he was made out to be.
From Car/Bike chases to smashing through windows of a major gang's headquarters, there is really no cooler game you can develop in the Mass Effect universe.

Some thoughts based on what I have read in this thread and the "smaller story" thread. Has anyone thought about Startrek: Voyageur as a model for a new storyline? Never been a big fan of the series or of Sci-Fi in general, but I thought the premise behind Voyageur was great. Relative ot the ME triology, a change of pace, smaller story but with lots of possibilities for stuff. I think the story has to begin with the end of ME3, because knowing the outcome of the series could really dampen the mood of the new game. Possible to link new story to the end of ME3 via what happened to the Mass Relays. Different galaxy or a part of the galaxy suddenly made inaccessible.
Refugee ship(s),new characters, (Shepard's retired or dead or otherwise occupied), multiple races. Maybe add some RPG/Action adventure gameplay in getting team together to confront new challenges. That is, have actual gameplay options that reflect the Paragon - Renegrade speach choices -- kill your way out or through or deal with potential antagonists with speach problem-solving stuff -- add to the gameplay possibilities w/o taking away from the core squad-cover-shooter gameplay.
Make it so?

heres my opinion. forget fans opinions and do your own thing really
i say this because no matter what fans say they will never be happy
i would agree with this but im kind of a hypacrit for doing so but, so long as its a full single player game and not an MMO and no type wasted on some sort of **** co-op mode like mass effect 3 then im fine with them doing there own thing. i may not like what they are doing but provided they dont go all MMORPG or FPS on us i dont care.

My wishes & suggestions:
- NO dumbed down First Person Shooter
- keep the balance between 3rd person action & RPG elements
- keep dialogue wheel
- less auto dialogue than in ME3
- new human main protagonist/player character (not Shepard)
- keep character customization (gender, overall apperance, skill-tree, etc.)
- keep ME2/ME3 combat system
- keep Paragon/Renegade system
- would prefer to see a game set after the events of ME3 [don't care if one of the ME3 endings would be declared canon to have a ME4]
- all new exciting storyline
- all new alien races and old alien races
- new enemies
- all new interesting NPCs & squadmates
- cameo appearances of ME1 - ME3 characters (if they survived previous games)
- all new planets & worlds to visit
- keep Galaxy exploration
- planetary exploration reintroduced (MAKO/Hammerhead or all new vehicle)
- more hub worlds/open world levels
- more Love Interest interaction/dialogue
- squadmates should agree/disagree with player's actions much more (like in DA2) and earn friendship/rivalry points (with the possibilty that they might leave your party with too much rivalry or special bonuses with enough friendship points)
- all new player's starship
- more starship customization options

You guys left us with this big cliffhanger. You can't leave us hanging like Caprica did!
Oh, and of course, I'd like the customization options to stay in tact, and maybe even let us pick a custom race.
And can we get an MMO with a gameplay style similar to Global Agenda? *is shot*
