I think the Tyranids from 40k may have been created first.

Trying to create some bigger and more dreadful enemy then Reapers would be bad idea. Mass Effect would end like Moonraker, when James Bond had to go father and father and suddenly we had space battles in Bond movie. But creators realised that and next Bond movie had classical spy plot about lost cipher device and series miracously cought new breath.

So new enemy shouldn't be necessary bigger threat then Reapers. Lets try something smaller and maybe evolve it during series. For example, when I remeber Talitha story from ME1 about batarian slavers, closer look on them should be quite scary too.

You don't need to top the Reapers.

Look at ME2 for instance. Suicide mission feels like saving the whole galaxy, when it's basically destroying only some insect-like goons.

Simple, everyone is now part cyborg in a galaxy were there still exists giant space squids that are known to hack into computer networks and indoctrinate beings by just being around them for a time. We find out the Reapers are actually harvesting us for knowledge and building materials and that now the process is easier than ever before. However, trying to bring this up to anyone in power is fruitless because they are enthralled by the Reapers. You must now construct a device to kill all the Reapers and nothing else from another set of ancient blueprints.

The Shepalyst from control would be a frightening opponent.

Synthesis solves nothing and adds a ton of new problems.....

In Destroy, you have tons of enemies who would make great antagonists, including those upstart Humans or even the Salarians.

You can certainly top them.

There can be terrors in the universe that makes the reapers look like little robots.

Create a true unknown, lovecraftian intergalactic or maybe even interdimensional nightmare.

OK for one the reapers spend most of their time sleeping. Outside the galaxy. They're not worried or bothered about threats from any other galaxy in the universe just ours.

Until maybe something comes along that's bigger and badder then them and makes them pee down their tentacles. :D :D :D
