Zevran, Shale, Oghren. These three have silly interactions with pretty much any other character you could throw into a party.
I usually have Leliana in my group, as she's my favorite among your rag-tag band of misfits... but her interactions are only ocassionally funny. Usually she takes a more serious stance even in conversation with sillier party members.

It depends a lot on the combination of companion you pick.
-You should always combine Oghren with a female partymember, preferably Wynne or Leli.
-Morri + Sten also gives nice banter, they definitely have some uhmmmm chemistry going, lol
-Morri also has some interresting discussions with both Leli and Alistair. Mostly due to Morri being their opposite
-Alistair + Leli + Wynne gives a comfy happy family feeling, only the kittens and the white picket fence is missing here. They're making jokes of each other, but always in some joyful way.
-Zevran and Shale I find more focussed towards the PC than towards bantering. They chip in, but are usually more fun in conversation with the PC
As for gameplay.. That's a tough one, but I'd say, try out a different setup for your party on every playthrough. One with as many mages as you can get, one without any mages, some with only 1 or 2 mages. Also try the different mage specs, a spirit or entropy mage plays entirely different from an elemental one. Same applies to warriors: to sword & board or not to sword & board. A party without a warrior at all makes for a challenge as well. You can do it with a rogue for example, but agro will be slightly harder to maintain.
In future, I also plan to make an opposite team, where members purposely won't agree over choices, and not use ANY gifts (apart from certain plot items) to keep them happy. I think it might be a challenge, to keep them together that way.

My first playthrough went well without Wynne or anyone else using healing spells I must say. She makes certain things easier to handle, that much is true, but not a single character is a "must".rafoca wrote...

truly I have zero interest in "effectiveness", just which companions are going to be the most interesting to talk to and learn about.
'hot' is not a deciding factor either, i'm afraid.

She is more of a "must" than other characters though. Sten can be replaced by Oghren, for example. Leliana with Zevran.
Sure you "could" make Morrigan your healer, but then you'd be lacking offensive spellpower.

Shale is cool. Interesting unit. Cool lines. Funny.
Zevran is amusing also. He can be allowed to be a fairy with a silly accent, because he's got some fun lines.

Last game it was Alister, Morrigan, Wynne.
This time through it's, mostly, Oghren, Sten, Morrigan.

Wynne. Her borderline-fatalism makes her an interesting twist on the straightforward kind-teacher trope.
Shale. A little one-note, but that one joke is good enough to merit some repeating, and there is a neat side-quest.
Sten. Mostly in that he provides insight into the most interesting and alien culture in the DA setting.
Al and Morrigan really didn't do it for me. Mostly because the game seemed to be trying too hard to get the player to like them (a cheezy way to make certain late-game story decisions more difficult for the player).

Oghren - Nothing bad about beer-swilling, red-haired, angry dwarves with big two-handed axes.
Sten - Trying to impress the token Borg guy is fun.

Alastair - He's a funny guy and often disapproves of my choices (Redcliffe) so I have to pay a lot of mind to how he will react.
Morrigan - It's hard not to like someone so sarcastic, direct and yet conflicted (however slightly). Her dialogue and options are well written and good fun.
Wynne - So far she provides good conversations on the nature of the unfolding events. Gives you something to ponder.

morrigan and alistair are fun together.
Leiana has been a constant in my party for 2 playthroughs now because shes the only rouge that has a good start on lockpicking, so i dont really know about a third

I like Leliana's personality, she's always my romance option when I create a character.
And Sten because even when he is in a good mood he is purpetually looking like ">=[" which is hilarious.

alistair is whiny and i dont understand how he can be a warrior at all really. morrigan and leliana are both there for your personal e-sex needs which makes them sort of trashy to me. oghren is just an idiot, and zev is gay and im not.

Alistair is always funny. Morrigan too. but those two are obvious.
I might give Zevran a try on my next next playthrough.
