Kate Beckinsale? Milla Jovovich? Jennifer Lawrence? Not saying that they should fill the shoes of Shepard, at all, but they certainly pull box-office numbers.
In worldwide ticket sales, most of the Beckinsale and Jovovich films typically make back three times their production budget. It's impossible to discuss The Hunger Games, given its rampant pop-culture fandom, but that one did very, very well with a female in the lead.
With the right writers and the right female lead, it'd do just fine.

Flip through these and check out the "Worldwide" figure: Resident Evil; Underworld.
Usually, ticket sales are between two- and three-times the production budget. That's not a bomb, even if it's not the same as whatever flavor-of-the-month science-fiction flick is tearing through the box office, such as the dreck that was Avatar. And though they're entertaining, none of the RE or Underworld flicks are well-written, yet they still do fine enough to keep making them. Prometheus did well too, and the lead was Noomi Rapace.
Decent writing around a strong-enough concept and a decent director could potentially make a successful FemShep Mass Effect film, even though it'll never happen given the insistence of the promotional materials and brand recognition. Nothing's set in stone without the write filmmaking essentials, though.
