They may try to make a game set either during (like the gap between ME1 and ME2, with a plot centered around the collectors) or before the ME trilogy.
My best bet if they make a game is that they won't make it single player, but rather co-op based. I can really see them experimenting with the MP right now.
But still, the more likely option is that ME3 was a franchise killer. (Check tv tropes)

Although, I could be swayed if ME4 has a short prologue featuring Shepard, (assuming survival, which I do) with some actual closure. i.e. He's living happily with [PREFERRED LI HERE] and had lots of little [PREFERRED CHILD COLOUR] children.

ME4, The Search for Shepard.
Destroy: After a time period where Shep is preserved, bring him back ala Lazarus.
Control: Many years in the future, his/her essence is data mined, consolidated and downloaded into a biomechanical body. (BTW it brings up all kinds of philisophical questions about what constitutes a soul).
Synthesis: Tough one since Shep's essence is spread across the Galaxy in a new form. Any ideas?
Denial (shooting the Catalyst): Peace loving advanced lifeforms with no real concept of war come across Liara's "message in a bottle" that also has some of of Sheps DNA lingering inside grow a new him/her (remember, DNA has a memory) just in time to save them.
Just some funny thoughts.

Sequel or side-qual seems most likely. There are a lot of subplots and backstories for the ME team to delve into. You could play as Garrus or Liara or more likely a new character, and you'd probably get to meet some of the classic ME characters during gameplay.Bester76 wrote...

If they left the current generation altogether, setting could be anywhere, and protagonist could be anyone. I don't think it'll be Shepard. They'd have to do a pretty big turnaround for that. Which is possible, but unlikely. Pressure would be huge to come up with a pretty good story to continue the Shepard saga if they did that. With a real chance for it to backfire.

Unless they are blue or somehow half Quarian or Turian (neat trick but anything's possible).
And anything Shepard-related would have to have a clearly defined single ending for ME3 or in most cases reaper drones or green "eyed" reapers are flying around serving tea.

Not for me. "No Mass Effect without Shepard" is my new motto. Old motto? Post-ending motto! (If they had properly ended ME3, I'd have been able to move on and play a new protagonist. Now I can't.)
