Act 1, 2 or 3? Does it even make a difference?
When's the best time to play the DLC?

It can. When you play it has an impact on dialogue; for example, I played Mark of the Assassin in Act 3, so everyone referred to Hawke as the Champion and Tallis recognized Hawke's status as basalit-an. There's also some different dialogue in Legacy depending on whether you play it before or after Leandra's death. Also, if you bring your love interest, pertinent romantic dialogue will only occur if you've actually shacked up; I waited until after asking Merrill to move in to play Legacy so I could bring her along.
As well, the loot corresponds with your level. I wanted to use Hawke's Key against the Arishok, so I played Legacy fairly late in Act 2. I played Mark of the Assassin just before the climax in Act 3 so that I'd have good armour and a new axe to use through the end-game.

Most of it makes the most sense if you play it in Act III.

There are different reasons to play the dlc. Gameplay wise, it is better to do it around act 2 or early 3, so that you get some use of the items for the rest of the game. I assume you are more interested in roleplaying reasons, if so, this is what i have tried.
Mark of the assassin:
Act 1 : best motivation for hawke to accept the mission (wanting money), best moment to do if you are going to be anti-qunari, otherwise your Hawke looks way too stupid. It also makes more sense that they call Hawke a fereldan and not a marcher (you´ve only recently arrived to kirkwall, so i feel the fereldan status is still relevant at that point in time). Hawke is treated as an amell, but with a fereldan father, to me it makes sense for the orlesians to look down upon hawke at this time.
Act 2 is pointless, it is the same as act1 but with (imo) worse justification and dialogues.
Act 3 is the alternative if you want all the dialogues (like LI dialogue), plus Hawke is treated as champion of kirkwall. This works well if you were friend of the arishok as tallis mentions your relation with it, it makes more sense for a qunari friend to continue helping her after the reveal.
Legacy:
Act 1 is nice enough, it fits quite good here if you want the whole family reunion. Personally i find the deep roads have enough darkspawn for one act, so i leave it for later.
Act 2 is decent if you are interested in doing it before mother dies, otherwise it isn´t really good. You get to see your sibling evolve a bit however.
Act 3 is in my opinion the best, as you get most dialogues and a break from a otherwise boring act. I also prefer the conversation with "dead mother" as i think it is more emotive. You also see how the sibling changes in personality since last time you saw him/her.
There is no hard rule as to which act is best, it depends on your hawke. For example, my hawke is anti-qunari, so the only way to do Mark of the assassin that makes any sense in his story is to do it in act 1 before the deep roads, otherwise there is a huge disconnection.
You have think where it fits best in your hawke story to go on that mission, as in for example the motivation: does it makes sense to you for your champion of kirkwall to want to steal from some random orlesian? Perhaps, if your hawke still feels Fereldan, and maybe is doing it for payback. If you can come up with a believable reason for your hawke to engage in that quest, then it is porbably a good moment to do the mission.
