Here are the important specs I can think of:
Windows Vista-64 bit
Can't remember processor right now, but it was top of the line in 2009 (and I have no problem with Skyrim, Dishonored, Witcher 2 at almost max settings).
XFX HD 7970 GHZ edition video card
50 gigs of harddrive space
8 gigs of ram
UAC is off.
Hardcopy version of Mass Effect 3.
TL/DR: ME3 won't initialize whether from a downloaded version or the hardcopy version. I get no Origin error message when I try to load the game from Origin. Repairing the install has not worked. Sometimes I get a game update error, sometimes I don't when I repair the install. Deleting the installers has not worked. I have the latest version of Origin, I think (the beta checkbox is checked). Loading it outside of Origin gives me the following error message: WINDOWS CANNOT ACCESS THE SPECIFIED DEVICE, PATH OR FILE. YOU MAY NOT HAVE THE APPROPRIATE PERMISSION TO ACCESS THE ITEM.
Back in July, I bought it online, and the game would not initialize. After messing with it for a week, I gave up and asked for my money back (took a while, but they relented and gave me a refund). Then I went Target and bought a hardcopy to see if that would work. It didn't, so I gave up until now, hoping that there would be some patches to origin or something.
So I reinstalled it last weekend, and I ended up having to jump through some additional hoops because Origin somehow forgot that my hardcopy version of the game was registered to my Origin Account. I even had to send them a picture of my signature, the date, and my manual with the promo code.
Tried to install it from the disc, and I got a disc read error prompt from origin, and it started to download. I said, OK, and let it download overnight, and I still had the same problem from all those months ago--it wouldn't initialize after clicking on the game, and repairing the install didn't help either. I uninstalled and tried another manual install from the disc, and this time it successfully read the discs. I thought I was in luck, but I ended up having the same exact problem. Also, I can't initialize it outside of origin either. I always get a windows prompt that says I don't have the right permissions.
Any help would be appreciated. Otherwise, I may have to get it on PS3, even though I've got my ME1 and ME2 saves on PC.
Mass Effect 3 in won't initialize. Bought it in July, never been able to play it.

Well, I haven't thought about that yet, so I'll try working on it when I get home. Do I need to do anything with the ports or just turn off the firewall completely or something?

you have ot create specific rules for me3, origin and eacore as allowed full access
not sure how you do that in windows default firewall but most programs include a feature that allow you to do this and all decent firewall products are windows certified so when there installed they take controll of the built in firewall and do this automatically.
if your useing an off the shelf product like mcafee or norton you need ot change the default behaviour from automatic handling and make it ask you (annoying popups) for everything
switching off a firewall program wont be enough in itself to allow the game to have internet access (the windows defualt setting is to disable everything not already granted access through allow rules)
hopefully that will help a bit, also there was a sticky thread somewhere about installation problems and how to "work around" various security programs which cause problems
if all else fails google is your freind
