Do mages have to have other mages in there blood line to have the gift (or Curse) I know the Amell line is strong in blood magic and that means there is a high chance when Leandra and Malcolm Hawke will produce a Mage child. But can mages be born with no prior connection? If you killed all the mage blood lines would there be no mages?

I've always thought about it like if your parents or grandparents were mages you had a higher chance of being a mage. A diathesis for magic if you will. But at the same time that the ability could manifest completely randomly.
After all magic is part of the WORLD of Thedas, it's not something outside it or unnatural, so no, I think that even if you managed to kill ALL the mages and ALL the mage bloodlines there would still be mages born.

It seems to be hereditary in the same way many characteristics such as hair and eye color and complexion are with dominant and recessive genes, e.g. a mage couple would be very likely to produce mage offspring, but it's also possible for it to skip generations or manifest under just the right combination of genes, so two people with little or no known 'mage blood' could have a magical child.
I don't think the rules so to speak have ever been explicitly laid down. I suspect most Thedosians believe it comes from the Maker for reasons unknown.

This is possible, it could really be a curse from the maker afterall.

Seeing as the number of mages in Thedas is rising at a fast rate, I'd say its less to do with bloodlines and something more mystical.

Actually, I expect it is entirely to do with "bloodlines"--seeing how blood in Thedas has some kind of strange mystical force. I expect it isn't, or isn't quite, genetic, however.

"Did you know back in Tevinter, there was a magister who made a research on the occurrence of magic ability through a bloodline?" ...well, that's little conclusive
but she somehow indicated that he found something
wasn't it also mentioned that isoldes grandfather was a mage and she had kept it a secret?
rhys is a mage as is wynne and it is possible i misinterpret it but it seemed to me like he was send from orphanage to tower without them having any prove of his magical ability... at least nothing's mentioned
I'd compare it with something between being lefthanded and eyecolor, only more uncommon and infrequent...
a 30% chance per parent and a all or nothing thing would be nice
it kinda bothers me that amell mages are (always) strong,
it'd be better if even nonmages can get exceptional strong mage children
