I'm going to start a new playthrough of ME1-ME3 and wanted advice on choosing the most interesting background for my new Commander Shepard. Spacer, Colonist, or Earthborn? Ruthless, War Hero, or Sole Survivor? What is the most interesting combination?

"Most interesting" is subjective.
If you want to try something new...then just try something new.
I only ever use one background...so I can't really help you...except to say that Earthborn, War Hero is always great, in my opinion.
Though I also headcanon a certain "Project Vitality" into my Shepards background...which makes it even better.

lol yes "subjective" but that's why I was asking out of curiosity of what some other players thought was "interesting".

I had done alot of Colonist/Spacers Ruthless/Sole Survivor backgrans in the past. My Soldier play through I just got into ME3 was an Earthbond War Hero. I did this so he would have more reason to want to save earth. Plan to have him and Vega in the final misssion ripping up the bad guys as they save the world. It's allso my almost all human play through, which is messed up until I can get Ashley back in the party so EDI is my alt for her. Though I'm thinking about doing an Engineer play through while I"m offshore (can't play my ME3 DLC while off line) and was going to go Spacer Sole Survivor....think of Aliens movie type stuff....

Each background has a different mission in ME1, in turn that leads to since minor differences later in emails our overheard news reports. Think that's about it.

I think Colonist/Ruthless is a good combination RP-wise, and those two backgrounds have my favorite ME1 tie-in quests.
Personally, I've played Earthborn/Sole Survivor (the default background) and Spacer/War Hero. I like the first a little better, but all the little extra touches with Hannah Shepard are nice in Spacer. It's a refreshing change of pace to have a PC who ISN'T an orphan for once.
