We already know what happens in the Rachni Wars, Krogan Rebellions, and First Contact War. To keep the series interesting they also have to keep the timeline moving forward.
A prequel setting might work for a four to six hour DLC or a full expansion, but it wouldn't work for an entire game IMO.

This is why I hate the Star Wars prequels...because it's too hard to suspend disbelief when everything looks so better, more crisp and new than what it is supposed to predate.

You don't know how a new sequel ends til you're at the ending; that sense of discovery in the story is what I want.

But ultimately I would prefer a brand new trilogy, that remains in the ME universe, eventually in a new galaxy (not sure about this but that's a possibility), which would have no reference to the conflicts, story plots and characters from the previous games, and therefore no save import.

And will people please STFU about canonizing the ME3 ending, Bioware knew there would be another Mass Effect game just not with Shepard, if they were aware of that they'd know how to work with all 3 endings and hey that gives us at least 3 replays of ME4, stop selling Bioware short they made 8 different storylines in the Old Republic I'm more than sure they can handle 3

The further forward you go, though, the less of a problem this becomes. Move forward 500 years and it'd be easier to swallow. There's probably even a sweetspot, somewhere far enough forward that it'll make sense that it's forged enough of its own new history to string a new story out of, but close enough that you'll still feel like Shepard made a difference. Hey, who knows, maybe Wrex and Liara might still be alive!
Personally? I'd circumvent that mess altogether, so I've voted prequel. Prequel makes more sense to me - plenty of stories to be told in the galaxy's past. "Pre-humanity" sounds... exciting. But! It's not my story to tell. As long as it's a new Mass Effect game, I'll be happy.

It's the fourth Mass Effect game, so that's what we've been calling it, but it's not been announced as a title, has it? Dragon Age 3 was only called Inquisition within the last couple of months.

Make it so you do the last mission of the game twice. The very first will be the same as the last, the only difference is the graphics should be much nicer the first time you do said mission. When you do it again at the end of the game, the graphics quality should be worse. Maybe the same as the first KOTOR.
