Missing a back pack when you lose an area does that mean you cant get 120 slots anymore?

If we're talking about the same one from the beginning of the game, yes. Once you're (forced) to leave there, you can't get back.

There are others to be had, but if you miss that one from the beginning you'll finish up with 110 of a possible 120 available backpack space.

Check back with stores after you finish the area, they will have new stuff, like backpacks.
