On my current playthrough, 2nd time through as a city elf female, my character fell pretty hard for Nelaros.  I am not sure yet if she will succumb to either Alistair's or Zevran's charms, but in any case am planning for her to be heartbroken and distant for a good while into the game.

My question is, if you reject the boys' advances, do you get an option later to change your mind or do those dialogue options disappear?

If you are clear about not being interested then you wont get another chance. I am not sure if there is an option that does not advance the romance but doesn't block it completly either. A no is a no though.

If all else fails, you can use a command in the developer console to set relations between companions, including starting a romance
