Definitely. My bread and butter is the Fury, but I can use a Ghost, Turian Sentinel, Huntress, and Valkyrie on occasion. I'm trying to expand on the other kits as well, but don't ask me to play gold with any of them yet, haha.

I try to play almost anything and my favourites keep changing. I like trying out different stuff, so I'm not particularly good at any class. =)

I have a lot of characters you could say I "specialize" with. I play pretty much all characters and switch each game typically so I can play well with any of them.

I don't get why people would limit themeselves in using 1 or 2 kits.. I think the game will get repetitive and boring quickly that way plus the game is not that hard, its not like Street Fighter where you need to specialize in order to be in a competetive level..
EDIT: After I went through the first page and actuallly read the OP, I think the title is a little misleading, spending more time with a kit does not automatically means that u specialize in it, it just simply means that its a kit that you are having fun the most. I guess "specialize" is too strong of a word to describe what the op means..
To answer the OP: I play a lot of kits, my choices on a mp session depends on my mood, and there are lots of times that i wished that there's a random choice..

N7 Destroyer, and Female Quarians.
Kinda enjoying the Valkyrie and Shadow, as well, but mostly stick to Destroyer, because I'M the main source of offense with the group I usually play with, so we suffer if i'm not Destroying enemies.

Specialized? Not really. But I'm better with Tech than Biotics.
I can play fairly well with most kits.
But I like to think I'm good with my favs.

I play a wide variety yet find myself never letting go of the good old Drellguard, Drelldept for when I want to use warp ammo and more dodging, Asari Valkyrie to melt everything, N7 Destroyer for the dakka, Turian Sentinel and N7 Fury. Most of them use Claymore, that's a lie I slapped a claymoar on all them at one point.

90% of the game, including the salarian and geth infiltrators, the kroguard, all the other classes I don't really like...
I'll stick with the set-ups that I enjoy, occasionally trying something troll-ish if I'm extra bored.

The challenges has definitely pushed me through a much larger variety of classes than I normally would play (which is just about everything now).
With the 200 waves, I did find that if I played only 2 or 3 characters while completing them, I got very proficient with them, but when bouncing around between characters, I've noticed I dont exploit each characters perks as well as I'd like, and usually a second game in a row is much better than the first.
While the 200 waves is a bit daunting at times, I do find that I've come to appreciate far more characters than I did originally, so I dont think I'll ever go back to specialization of any particular characters.
The ammo/tech powers explosion change REALLY helped a lot of classes as well, making many far more enjoyable and competitive.

I feel that part of the whole appeal of ME is personal customisation, so it would seem perfectly acceptable to specialise in a single character.
That said, you'll be missing out on like 90% of the game, not to mention fun! So no, I don't specialise. Preferences? Of course. But no specific specialisation.
Turians, Quarians, and Geth (though oddly not the GI) are my go-to aliens, but Humans never let me down, either.

I have several characters I really like (Shadow, Turian Sentinel, Asari Adept, Batarian Soldier,...), but the one I absolutely love is the Salarian Infiltrator (with my Valiant still at II
). I've been doing the challenges since Retaliation came out and almost never play him anymore, but just today I took him out for a spin once more and fell all over in love again. <3
