I'm about to embark on what will most likely be my last ME1-3 playthrough and when I reach ME3 I want to focus on melee combat.  I know it won't be efficient, and quite possibly not even feasible for encounters with Brutes and Bashees.  That being said, I'd like opinions on what would be the best base class/bonus power.  Here's my thoughts:

class:  Infiltrator or Vanguard (either cloak up beind the target and heavy melee or biotic charge back and forth)

-  I like the idea of cloak as its the ultimate defensive skill in addition to being tactically useful.  Though, biotic charge with +50% barrier recharge is appealing for when I'm "caught".

-  I don't want the playthrough to completely stall if melee isn't working on a boss (Brute/Banshee/Mech), so I'm leaning to Shotgun as a backup since I've played with all other weapon types extensively.  Infiltrator would get a bonus while cloaked, but Vanguard gets a full time 15%.

Bonus power:  Fortification or Stasis

- Fortification would grant greater DR and, upon purge, a much stronger melee bonus.  Stasis with bubble evolution would give me greater crowd control while I try to pick off enemies one by one with my fists.

Note:  I would not be opposed to using squadmate powers to strip defenses or provide supplementary crowd control, but I would be attempting to kill most minion level npcs with melee attacks to the extent it remains enjoyable/feasible (I don't plan on punching down many turrets).

Did I leave any strong choices out?  The idea of synerging the fitness tree talents that I've never used, with weapon mods I've never used and armor bonuses that I've never used is appealing, which is why I'm going down this masochistic path :)

Any thoughts on class/power/weapon/squadmates to help me out?

Infiltrator with Fortification may be your best bet. Here is a melee-focused Quarian Female Engineer build in multiplayer.

If the melee mechanics work the same way in single player for the Infiltrator as they do in multiplayer for the Quarian Female Infiltrator (QFI), then you are more than capable of a melee-only build.  Since the melee is the same (electrical slash), you should be able to get around the same bonuses as the QFI:
 - all melee damage in the Fitness talent tree (Same as multiplayer, +95%?)
 - Cloak with Damage (+90% for single player) and Melee (I think it is the same as multiplayer, +50%)
 - Up to +50% melee damage from armor
 - extra melee damage from Fortification when purged for a limited duration (20 seconds? Do not know offhand)
 - +100% vulnerability from Sabotage with Tech Vulnerability (+50% more than multiplayer)
 - melee damage mods for weapons. I think one of the Omega DLC shotgun mods gives you more melee damage than the regular shotgun melee mod (+50%)

Melee Focused Sentinel

Tech Armour:
 Durability, Melee Damage, Durability
Fitness: Melee Damage, Shield Recharge, Melee Synergy
Fortification: Melee Damage, Shield Recharge, Durability
Offensive Mastery: Rank 5 should be weight capacity.

The rest of your power evolutions are up to you.

How to play:

Carry a light shotgun.

Activate and purge fortification on a loop. That way you'll get the important melee damage bonus.
Keep using your heavy melee attacks.

Make the most of grab kills whenever you can.

Avoid using your melee attacks on any enemies with sync kills (Atlas Primes, Phantoms, Banshees and Brutes).
When using your melee is a bad idea, your powers and squadmate powers will handle the situation.

Fortification Infiltrator w. Sabotage will likely do the most melee damage.

Sentinel w. Fortification and Throw might be more fun.

Melee Focused Vanguard

Biotic Charge: Spec for radius so that you can stagger as many foes as possible. The other evolutions are up to you.

Fitness: Melee Damage, Melee Spree, Melee Synergy

Fortification: Melee Damage, Shield Recharge Speed, Durability.

How to play:

Charge, follow up with heavy melee.
On enemies with more hit points purge your fortification for the melee bonus and unleash havok.

Nova is recommended as are shotguns (light shotguns).

Keep your loadout light, since you'll be using biotic charge and fortification a lot.
Also take full advantage of grab kills.

Avoid using biotic charge and melee attacks on Banshees, Brutes, Atlas Primes and Phantoms.
When  melee and charge are poor choices, use your guns and powers.

this thread makes me wish we could play as a Krogan or a Slayer in SP.

Thanks everyone for the input!  Just for clarification on sabotage, is the 100% vulnerability to tech damage in any way applied to melee attacks from an infiltrator or only other tech attacks such as incinerate?  I assumed melee damage was a separate classification of damage (other than tech or biotic).

Vanguard, they are kings of Close Combat. You need AoE charge and full barriers refill evolutions btw.

According to the thread I linked to earlier, the Tech Vulnerability of Sabotage is apparently being applied to the Infiltrator's melee (the QFI has the same melee as Infiltrator Shepard). Either it is a bug or some melees are considered "tech" like the Infiltrator's melee. The Tech Vulnerabiltit also applies to all tech powers, including the backfire of Sabotage itself.

I say Vanguard because you have charge (more like a body slam melee), a great biotic punch (the VG heavy punch), and Nova. Melee away!

I have an interesting vanguard melee-based class I've been using lately. The vanguard is unique and he/she has two heavy melee attacks: one standing heavy melee and one that is similar to nova that activates when you hold down the heavy melee button while sprinting. I don't substitute nova for heavy melee attacks like some people like to do, I just use nova to stagger enemies and as a kind of extra melee attack. This class is great for melee as powers can be used as an extension of melee attacks.

Powers:

Cryo ammo: -Rank 4: Duration
-5: Headshot damage
-6: Damage combo
Although it has nothing to do with melee attacks, you're going to want to use a shotgun for this class so having the Cyro ammo softens enemies up, weakens armor and makes them slower so they're easier to melee.

Pull: I'm not going to go through this one simply because it isn't really important unless you want to pull enemies close for a melee attack or shotgun class.

Biotic charge: -4: Radius
-5: Weapon synergy
-6: Barrier
I use biotic charge to stagger multiple enemies so they can be dispatched with melee and shotguns without too much trouble. You can go for power damage at rank 5 but the fun of this class is in the melee attacks so you don't want to make your powers too strong.

Shockwave: -4: Impact radius
-5: Reach
-6: Recharge speed
Shockwaves is usually a generally useless power as it's not quite as effective as it was in ME2 and the core powers for a vanguard, charge and nova, are good enough. However, for this class shockwave is very useful as a staggering melee attack and the reach upgrade at rank 5 means you can stagger enemies in the distance while you melee closer ones. It can also pass through cover and allow you to rush them.

Nova: -4: Force and damage
-5: Half blast
-6: Sustain
Nova is useful for staggering enemies and softening them up for melee attacks. You're going to want to have a damage boost either at rank 4 or 6 so you can take out sheilds and barriers easier.

Assault mastery: Put all points into weapon damage and power damage upgrades and weight capacity increase.

Fitness: -4: Melee damage
-5: Melee spree
-6: Durability

Bonus power: Use either defense matrix or fortification. I personally use fortification because of the melee damage bonus but if you want better shields then use defense matrix.

Fortification: -4: Melee damage
-5: Recharge speed
-6: Power Recharge

Weapons: As I mentioned earlier, shotguns work best with this class. You will want to use a fairly light shotgun because power recharge speed is fairly important. I personally use the AT-12 Raider from the leviathan dlc because it is quite light when upgraded, has a good fire rate and does as much damage as a claymore or spike thrower when the right attachments are used.

Armor: This down to personal preference but I like to have melee based armor for this class.

Helmet: Death mask
Chest: Ariake Technologies
Shoulders: Kassa Fabrication
Arms: Kassa Fabrication
Legs: Ariake Technologies

I hope this class is appealing to people because although it isn't entirely melee based, I tried to make it as easy and fun as possible to play as this class even on the hardest difficulties.

Not bad.  I would just caution PC players that Radius on Shockwave can sometimes cause it to bug out.

I am a console player so was unaware of this so thanks for the extra info. I'll also add that shockwave works well with liara's warp for setting up biotic detonations so you'll be able to do some good damage at close range to armoured enemies like brutes without them insta killing you.

For a melee game, sure.  I did a Cryo Sentinel that was sort of CQB based not long ago, and did a Shockware Adept way back when.  This is sort of a mix of those two.  I have some missions to finish up with an Engineer currently though.

How did you spec your class for the CQB sentinel? I've been looking for way to make my sentinel playthrough fun.

I didn't try to go for pure melee or anything on that guy, it was mainly to have some fun with Tech Armor and Cryo Blast.

TA - Damage and Radius, Power Synergy, Durability
OM - Force and Damage, Weight Capacity, Force and Damage
Fitness - Durability, Shield Recharge, Durability
Fortification - Durability, Power Synergy, Durability

You can pick whatever other powers you want to go with it.

I do not have the RL reflexes for melee, so Vanguard is not for me. Plus spamming the 'F' key sounds somewhat dull.

And Soldier is too repetitive for me; not enough variety and choice other than ammo.

The remaining have all been quite entertaining, so there is no best choice; depends on what you enjoy.
