I think I did it on either Hardcore or Insanity. After my first few playthroughs, I made the jump to Insanity. I had Wrex and Grunt with me when I beat the Mirror Challenge, and despite what I've seen people say, I could only ever do it with them (made sure to give them Claymores too)

I can get past Round 1 quite easily, It's round 2 i can't get past because of the Vanguard enemies. Charge+Nova is a nightmare.

I have the same issue. Once I got to the 3rd round, but ended up getting swarmed. The Soldiers and Engineers are the easiest to deal with, as are the Adepts. The Infiltrators are easy to take down as well, if you can get them pinned long enough. Sentinels are a bit more difficult to take down due to their increased damage resistance and shotgun, but enough focused fire should handle them.
The Vanguards are the hardest simply because they kept Charging away from you when you take down their shields. I usually save them for last. And I suggest grabbing Energy Drain. Can be really helpful in a tight spot.
Have you ever continued an argument on BSN for at least a week?

Never really used Defense Matrix myself. In my last Vanguard playthrough of ME3, I just spammed Biotic Charge and Nova in every fight. Think like the Novaguard build for ME3 MP. A lot of the tactics I use in the SP I developed in ME3 MP. Well, all except my Claymore-wielding Sentinel Shep. That was the first time I did a Shotgun Sentinel build.
