From what I understand, no. Then again, I haven't played any BioShock games yet; this is just what I've heard.

Theres a slight nod the the first game at some point in the game but nothing intrinsic.
The theme of personal choice is a big thing in the game, which is related to the first one but thats about it. No big daddies, splicers or murdering little girls.

As already stated, not really. There are a few direct connections between the two but nothing major. You should have no problem understanding the plot without any real knowledge of the first Bioshock.
