Got it back in '10. That's also the last time I played MGS4.
But... IIRC, the best way to do it is just to tranq everyone that the Rat Patrol don't cap themselves, and just stay low and camo with the environment to ensure you don't die, seeing as how you're low on rations.

I recall that I couldn't get past Crying Wolf for some reason; she kept spotting me.
The practice run took just over 5 hours. About 5: 48 or something. To get the Big Boss emblem you need to complete it in less than 5 hours w/o killing any bosses (you're allowed to kill Vamp fyi, but not the B&B unit bosses).
The actual run took me about 3 & a ½ hours.

I did the same but it took me about 4 hours 5 minutes. One of the few games I cared about enough to get the highest score. I've considered doing another few runs because of the trophy patch though.

From what I recall, the assault on the compound with Meryl was the toughest part for me, along with the motorcycle chase in Act 3.

Especially if you're not killing anyone. Otherwise, it's a nice bit of fanservice to MGS3 fans.
