I was wondering if a Soldier can also be built by ignoring the two cooldown-based powers, A.R. and C.S., and picking a defensive power (D.M. or Fortification) so I could use an extremely heavy loadout (-200%).
Compared to the Tank Sentinel, the Tank Soldier would "waste" only two powers instead of three (C.B., Warp and Overload), would be much more effective than the tank sentinel on setting up power combos (thanks to the three ammo powers) and would kill enemies faster thanks to the weapon damage bonuses. However the price would be increased fragility and lack of the good no-cooldown throw.
Thoughts?

The short answer is yes.
Fortification specced for maximum damage reduction (+40%) would mean that you would only take 100% of incoming damage when out of cover, as opposed to the 140% you would take without it. Fortification specced for Power Damage evolution at rank 5 would also increase your ammo power damage.
You will get more weapon damage than a tank Sentinel, as you could focus Combat Mastery on weapon damage for +25% weapon damage. That is in addition to any weapon damage bonuses you can get from Armor and Intel.
The issue that -200% cooldown that pretty much all classes have is limited detonation options. Grenades are limited, so you have a maximum of 4-6 Frag Grenades, depending on how you want to spec them. Certain missions seem to have more grenades than other missions, so you may find yourself unable to detonate your own explosions as often as you would like.
Liara would probably be your best squadmate. Singularity would be good for crowd control if you do not want to get shot (and Stasis can help as well). Warp can detonate explosions and you can get her Warp down to about 5.5 seconds cooldown.
If you want to prime explosions only, then Garrus and Ashley may be your next best squadmates with Concussive Shot on a maximum low cooldown of 6.5 seconds.
Now the one good benefit of not caring about cooldowns is that you can take whatever weapons you want so you can kill enemies with whatever weapons you want. At this point it is practically a straight-up shooter which is not necessarily a bad thing. You will be relying on your squadmates a little more than you would otherwise, but that is not necessarily a bad thing either.

I ran a basically CD-independent soldier (Revenant/Black Widow) and it worked out fine. Occasionally used AR to regain shields.
It was my first run actually (insanity).

It plays fine. It doesn't fit my style, personally, but it does play fine.
I'm not sure it would be better at explosions than the tank Sentinel, considering the Sent can detonate a biotic bomb every 2-3 seconds off of Liara's Singularity. If anything, it might be trading the raw power of the Sentinel's combos for the versatility of more types of combos. But, being based on ammo types rather than normal powers will make them weaker.
My recommendation for running like this would be to take Garrus and Liara as teammates. Use Incendiary ammo for Shep and Disruptor for the squad. The squad will be doing shield damage and CC with their shooting, complimenting their powers. Incendiary will make short work of armor, especially when debuffed by Warp and/or Proxy Mine.
If you want a more "all guns, all the time" approach, I like taking Garrus and James with a Soldier Shep. Same deal, IA for Shep and Disruptor for the squad. I specced James' grenades for anti-sheild and his Carnage for anti-armor. Shep's grenades where also anti-shield. Had no problems whatsoever handling defenses. In a New Game+ it works fantastically with the squaddies carrying Falcons.

Thanks for the tips, for the first time in ME3 I'll go for for Fortification instead of Defense Matrix, since (1) Fortification increases the damage of ammo powers, (2) Adrenaline Rush can work as an emergency shielding (3) using AR instead of DM increases lethality.
I've used Defense Matrix with all the other five classes.

You can ignore powers completely. There was a guy that used to post here frequently that used to run his soldier that way.
Take weapon damage evolutions in passive. Squad Incendiary with Explosive burst, and perhaps Squad Disruptor for the Rannoch arc.
It will not be quite as powerful as a character that spams Concussive Shot, or uses Adrenaline Rush regularly with a light but potent loadout, but it will be more than enough to get through the game.

Thanks. I've decided I'll use a mid-weight loadout composed by a variety of Assault Rifles and Shotguns. Therefore I'll have some cooldown-based powers, but thanks to the tips I'll use Fortification instead of the previously planned Defense Matrix.

You have a concept for a type of commando.
You can make it work.
Each of the 6 classes are very versatile.
I can't think of a single type of commando that you couldn't build with the right class, power evolutions and weapon loadout.
Cooldown independent tank soldiers are fantastic.
Cryo ammo for squad
Incendiary ammo for armor, health and organics.
Disruptor ammo for shields, barriers and synthetics.
Grenades for flushing out clusters of enemies.
Fortification for protection.
Adrenaline rush and concussive shot ditched.
Ditching adrenaline rush and concussive shot will put you at a disadvantage, but on the up side you're carrying every gun you want to carry.
Your favorite Pistol.
Your favorite Sub Machine Gun
Your favorite Shotgun
Your favorite Assault Rifle
Your favorite Sniper RIfle
Armor piercing mods might be handy. Or bring Javik or Liara to handle pesky cover abusing enemies.
