Honestly the first one.
Walking aboard the Normandy for the first time, the mass effect theme in the back ground, the Normandy prepping to jump through the relay, seeing your Shepard for the first time, and that string of energy crackling off the relay before it snaps you through the the relay for the first time. Blew me away.
Others:
Virmire in its entirety, I'm serious, favorite mission in the trilogy hands down

God, this is so hard...
Uhhh, i think I have to go with the romanced version of the conversation with Tali after getting peace on Rannoch, the dialogue there is just very touching a well done.

This may sound odd but the end of Mass Effect 3. I liked how the ending was handled I just didn't like the original ending with just different colors for the same outcome. The extended cut made the ending worth while for me. My 2nd favorite moment is when Jack gives you the tattoo during the Citadel DLC if you romance her. Jack is my fav character in the series and its awesome to see her get emotional and really care about Shepard and her students as well.

The first time Tali starts rambling when you begin the quest for her romance. In second I'd have to choose when Anderson walked into Udina's room and knocked him the **** out or the entire ending of Shepard vs Leng on Thessia. The first time I did that, it hurt so much.

but perhaps after everyone shows up after you killed the Clone , the End of an Era short scene.
Because it makes you so aware that this is the end of Mass Effect.

Honestly, I don't know if I could pick any one scene as a favorite. So many of the ones that come to mind are my favorites because of the amazing scenes that came before them. But, I guess I'd have to go for the origins: When I booted up ME1 for the first time, and my life was never the same.

For me it might be Shepard's speech upon taking command of the Normandy. I usually do the "long" version picking all the Paragon (or at least non-Renegade) options, and I find that it really captures my canon Shepard's philosophy about humanity's role in the galaxy and the need to defend others against threats like Saren. I'm a sucker for "make the world (or galaxy, in this case) a better place" narratives and this gave me the sense that Shepard's mission would be part of that while also dealing with the immediate threat.

1. Conversation with Sovereign. Never have I felt such impossible odds in a game before.
2. Battle of the Citadel. Truly epic scene.
3. End credits of ME1.
4. Destruction of the Normandy and revival of Shepard.
5. Suicide Mission
6. When Liara takes over as the Shadow Broker. I believe my exact thoughts were: "No way. No WAY! Awesome!!"
7. Both versions of Mordin succeeding at curing the Genophage ("I made a MISTAKE!" and "I am the very model of a -").
8. Aftermath of Thessia. Shepard is defeated for the first time, and Joker snaps.
9. "That was for Miranda you son of a b****!"
10. When the fleet comes through the Earth relay.
If I had a gun to my head and had to pick one though, it's probably be the Suicide Mission. The first time I played it was nerve-racking. Who do I send? Is this person going to die? I hope I don't screw up...And the MUSIC! Best in the series, imo.

It really is, and I ended up getting all of the ME2 albums along with some of 1 and 3. I think that a lot of what made me pumped up during the final act of Mass Effect 2 was really the music and how well it fit into the scenes, like when you have Shepard give a speech to the rear guard and subsequent departure on the platform and the end run to the Normandy. Lair of the Shadow Broker probably has some of my favorites too, but the suicide mission theme integrated with the gameplay is hard to beat.

Mass Effect: Being able to carry a save over into the sequal. A very cool feature which I'd never experienced before.
Mass Effect 2: The whole game. I've completed it so many times, that it feels like that game made specifically for me.
Mass Effect 3: Tough one between Wrex elbow dropping a car, and the synthesis ending.
