I think Garrus during his Archangel days was one everyone was pining for the most.

Another maybe Jack from breakout of Pragnia up to before she got boxed in Purgatory. Wrex during his mercenary days before the Normandy. Legion's expedition to find Shepard-Commander. Samara's hunt for Morinth. Zaeed's, Kasumi's Mordin's etc. lots to choose from really.

While I really love Kasumi, she's probably my favorite character besides Ashley, I don't think a spin-off would work for any character really.
There is a central protagonist for a reason. What usually attracts people to a story like Mass Effect is that protagonist, and not just any crew member.

Kirrahe. He was Commander Shepard before Commander Shepard. He was already on his Mass Effect 2 Suicide Mission when Shepard met him on Virmire. He probably gave my favorite ingame rallying speeches ever. I want to hear more about him holding the line.

Tough one. I can imagine a whole bunch of characters with many backstories that could make a decent game:

Garrus: Obviously being a C-Sec officer means a lot of police drama-style stories and situations. Rich pickings here.

Tali: Heard so much about the Quarians and the challenges and discrimination they faced. Might be interesting to walk in their shoes a bit.

Zaeed: As Joker said, he's basically Shepard who takes cheques. Kick ass and get paid for it? Yes please.

Wrex: Similar to Zaeed, I would think.

Samara: I wouldn't want to play as a Justicar - too many rigid rules - but it would be interesting to see how a Justicar's actions affects the world and people around her, and how they react back.

Jack: Grand Theft Auto in space, anyone?

Kasumi: Go on heists, evade cops and Spectres, find passionate love in Keiji, get heartbroken by Hock. Hell, even how that "Goto's Four" in the Citadel DLC turned out interests me.

Mordin Solus: Go on missions with the STG. Get into heated arguments with Kirrahe. Play singing mini-games.

Javik: He mentioned he once had a ship and crew just like Shepard. Playing through a constantly losing war might be too much of a downer, though.

Aethyta: Everything from her wisecracks, her headbutting a Krogan, to how the heck she got a squad of Asari commandos at her command tells me she's more badass than she lets on. I have to know.

That's just off the top of my head. There may be more.

I am hearing you. Someone posted the other day it'd be cool if they did a ME chronicles series where you get a few story options from the main menu. Imagine if they did 5 or 6 stories that were maybe 5-6 hours long each?

So all of those could work but rather than Javik maybe an older relative of his in a war with a different race, just before the reapers hit. 

And Captain Anderson, maybe a mission in the fcw and then his mission with Saren.
