With low level characters and on higher difficulties, RPG combat such as that of Dragon Age practically requires a tank in the party. This is why there will always be a tank companion introduced early into the party, in case that the player character is not a tank. The game would be imbalanced and not very enjoyable otherwise.
Moreover, this tank character will often stay with the party throughout the whole game, because without him/her the player often cannot have the traditional Tank - DPS - CC combo. In DA:O this early tank is Alistair. He joins you first and stays with you for the whole game unless you replace him with Loghain - this is to ensure you will not be without a tank. In DA2 this tank is Aveline. She also joins first and stays till the end of the game.

So it's safe to say DA3 will feature another tank character who joins your party early and stays till the end of the game. Who will it be?
The answer is quite easy. It is most likely Cassandra - she's a warrior with a steadfast (i.e. tanky) personality, and from the trailer she looks to be the new Aveline. Her character is relatively unexplored so it's easy to come up with reasons for her to stay with the Inquisitor till the end game (a secret mission from the Divine, for example).
This will also make it more likely for her to be an LI because there would be more time for her to interact with the PC. Aveline not being actually romanceable in DA2 have cause fans of her (and strong women in general) to be upset. This is the golden opportunity for BioWare to remedy that.
Another option is Cullen. He'd of course be an anti-Magic tank who will be very useful since you will likely fight a large number of demons in DA3. However, the problem is he has a pretty clear anti-Mage sentiment. If the Inquisitor is pro-Mage this may cause Cullen to leave the party. So Cullen is less likely than Cassandra to be the main/first tank of the party.

In addition to tank, a Rogue would also join the party early and stay till the end game, because without Rogues, traps and locks are a pain. The designers would not torture players who don't play as a Rogue with that for a very long time. This is one reason why Varric joins your party very early in DA2.
In DA3, you will still have a Rogue join your party early for the same reason. I hope it's Varric - and by the looks of the trailer it may well be. He's also one of the most beloved character BioWare has ever created so it would be a wise decision to include him again.
Moreover, the Inquisitor asking Varric to tell stories about Hawke/Warden is a great way of referencing the choices the player made in DA2/DAO, without introducing risky Hawke/Warden cameos which may be OOC (like Revan in SWTOR, ack). This also fits Varric's personality and allows for some great dialogue writing. So it's almost sure to happen.
So yeah, Cassandra and Varric fans should rejoice. You'd almost certainly see a lot of them in DA3.
